杀松材线虫细菌HEBAU PA-7鉴定及其发酵滤液稳定性分析OA
Identification of Bursaphelenchus xylophilus-killing bacterium HEBAU PA-7 and stability analysis of its fermentation filtrate
为鉴定松材线虫Bursaphelenchus xylophilus生防细菌HEBAU PA-7,通过形态学观察和分子生物学技术确定其分类地位,并采用直接触杀法评价其发酵滤液的杀线活性和稳定性.结果表明:菌株菌落黄绿色,圆形,大小不一,菌体杆状,有端生鞭毛;菌株与铜绿假单胞菌Pseudomonas aeruginosa亲缘关系最近,同源性高达99.93%;菌株发酵滤液对松材线虫具有较强的杀线活性,发酵滤液稀释10倍处理松材线虫72 h后,校正死亡率仍能达到100%,其杀线活性不受温度、蛋白酶K和紫外线的影响,但不耐强酸、强碱,最适pH值范围为6~8.菌株HEBAU PA-7发酵滤液具有较强的杀线活性和环境稳定性,将为该菌株的开发利用提供参考.
To identify the biocontrol bacteria strain HEBAU PA-7 against Bursaphelenchus xylophilus,its taxonomic status was determined by morphological observation and molecular biology techniques,and the nematicidal activity and stability of its fermentation filtrate were evaluated by direct contact method.The results indicated that the strain colonies were yellow-green,circular,and vary in size,with rod-shaped bacterial cells and polar flagella.HEBAU PA-7 was the most closely related to Pseudomonas aeruginosa,with a homology up to 99.93%.The fermentation filtrate of strain exhibited strong nematocidal activity against B.xylophilus,achieving a corrected mortality rate of 100%after 72 hours of treatment with a 10-fold dilution.The nematocidal activity of the fermentation filtrate was unaffected by temperature,proteinase K or ultraviolet radiation,but was sensitive to strong acids and alkalis,with an optimal pH value range of six to eight.The fermentation filtrate of strain HEBAU PA-7 demonstrated strong nematocidal activity and environmental stability,providing a reference for the development and utilization of this strain.
