不同体型双峰驼血浆外泌体的分离及其蛋白质组学分析OA
Screening for Differentially Expressed Proteins in Plasma Exosomes of Bactrian Camel with Different Body Types
为探究两种不同体型的双峰驼血浆外泌体在蛋白质表达方面的差异,本研究采用超速离心法分离双峰驼血浆外泌体,并利用透射电子显微镜(TEM)和纳米颗粒跟踪分析技术(NTA)对分离的外泌体进行形态学观察与鉴定.基于蛋白质组学技术,研究对消瘦体型与正常体型双峰驼的血浆外泌体进行了差异表达蛋白(DEPs)的筛选,并对DEPs进行GO功能注释、KEGG信号通路富集及亚细胞定位等生物信息学分析.结果显示,超速离心法分离的双峰驼血浆外泌体呈杯状或圆形形态,直径约为100 nm,总体尺寸分布范围为30~150 nm.利用串联质谱标签(TMT)结合液相色谱串联质谱(LC-MS/MS)技术共标记了4 903个肽段,筛选出172个DEPs.生物信息学分析结果表明,这些DEPs可能与脂质代谢相关的信号通路相关,并存在蛋白之间互作关系.本研究结果为进一步探讨双峰驼血浆外泌体的生物功能提供了新的方向和重要资料.
In order to investigate the differences in the protein expression of plasma exosomes from two different body types of bactri-an camels,this study used the ultracentrifugation method to isolate plasma exosomes from bactrian camels,transmission electron mi-croscopy(TEM)and nanoparticle tracking analysis(NTA)were used to observe and characterize the morphology of the isolated exo-somes.Based on the proteomics technology,plasma exosomes from wasting body type and normal body type bactrian camel were screened for differentially expressed proteins(DEPs),and bioinformatics analyses such as the GO functional annotation,KEGG sig-naling pathway enrichment analysis,and subcellular localization were performed on the DEPs.The results showed that the bactrian camel plasma exosomes isolated by ultracentrifugation had a cup-shaped or rounded morphology,with a diameter of about 100 nm and an overall size distribution ranging from 30 to 150 nm.A total of 4 903 peptides were labeled using the tandem mass spectrome-try tagging(TMT)combined with the liquid chromatography tandem mass spectrometry(LC-MS/MS),and 172 DEPs were screened out.Bioinformatics analysis showed that these DEPs may be associated with signaling pathways related to lipid metabolism and pro-tein interactions.The results of this study provided a new direction and important information to further explore the biological func-tions of plasma exosomes in bactrian camel.
