基于Ca2+膜电位调节作用探讨大豆卵磷脂对小鼠精子质膜完整性的影响OA
Effect of Soybean Lecithin on Plasma Membrane Integrity of Mouse Sperm via Its Regulatory Effect on Ca2+Membrane Potential
目的:通过加入不同剂量的大豆卵磷脂(Soybean Lecithin,SL),分析SL与精子质膜胆固醇含量之间的关系,在钙离子(Ca2+)膜电位调节作用的基础上,探讨SL对小鼠精子的冷冻保护效果.方法:将 40 只 8 周龄健康雄性小鼠取得的精子随机分为 5 组,分别为空白组(BC组)、模型组(MC组)、低剂量组(LD组)、中剂量组(MD组)、高剂量组(HD组),其中BC组不做任何处理,MC组只加入冷冻保护剂(CPA),其余 3 组分别加入含有不同浓度SL(5、10、15 g/L)的CPA,经冻存复苏后对各组精液冷冻保存后精子活力、活率、直线速率、曲线速率、胆固醇含量、钙离子(Ca2+)含量、精子质膜完整率、精子膜电位等指标,观察分析SL对小鼠精子的冷冻保护效果.结果:与BC组相比,MC组的精子活力、活率、直线速率、曲线速率、质膜完整率、Ca2+浓度、膜电位均有升高(P<0.05),总胆固醇(T-CHO)含量无差异(P>0.05).与MC组相比,MD组的精子活力、活率、直线速率、曲线速率、T-CHO含量、质膜完整率、Ca2+浓度、膜电位均有升高(P<0.05);LD组仅质膜完整率、Ca2+浓度、膜电位升高(P<0.05);HD组仅T-CHO含量升高(P<0.05).结论:10 g/L浓度下的SL可最佳程度地调节精子Ca2+内流、稳定膜电位来保护精子质膜完整性,从而提升精子质量,达到冷冻保护效果.
Objective:By adding different doses of soybean lecithin(SL),we analyzed the relationship between SL and cholester-ol content in the sperm plasma membrane,and explored the cryoprotective effect of SL on mouse sperm based on its regulatory effect on calcium ion(Ca2+)membrane potential.Methods:Sperm samples were collected from 40 healthy 8-week-old male mice and randomly divided into five groups:the blank control group(BC group),model control group(MC group),low-dose group(LD group),medi-um-dose group(MD group),and high-dose group(HD group).No treatment was administered to the BC group,while only cryopro-tective agent(CPA)was added to the MC group;the other three groups were treated with CPA containing different concentrations of SL(5 g/L,10 g/L,15 g/L),respectively.After cryopreservation and thawing,following indicators of sperm in each group were detected and analyzed:sperm motility,viability rate,straight-line velocity,curvilinear velocity,cholesterol content,Ca2+content,sperm plas-ma membrane integrity rate,and sperm membrane potential,and the cryoprotective effect of SL on mouse sperm was investigated.Results:Compared with the BC group,the MC group exhibited significant increases in sperm motility,viability rate,straight-line velocity,curvilinear velocity,plasma membrane integrity rate,Ca2+concentration and membrane potential(P<0.05),with no signifi-cant difference in T-CHO content(P>0.05).Compared with the MC group,the MD group showed significant increases in sperm mo-tility,viability rate,straight-line velocity,curvilinear velocity,T-CHO content,plasma membrane integrity rate,Ca2+concentration and membrane potential(P<0.05);only the plasma membrane integrity rate,Ca2+concentration and membrane potential were increased in the LD group(P<0.05);and only the T-CHO content was elevated in the HD group(P<0.05).Conclusion:SL at a concentration of 10 g/L can optimally regulate sperm Ca2+influx,stabilize membrane potential to protect the integrity of sperm plasma membrane,thereby improving sperm quality and achieving a cryoprotective effect.
