基于孟德尔随机化探究组织蛋白酶与急、慢性牙周炎的因果关联OA
Exploring the causal association between cathepsin and acute and chronic periodontitis based on Mendelian randomization
目的:本研究旨在利用孟德尔随机化(Mendelian randomization,MR)方法,探究9种组织蛋白酶(cathepsin,CTS)与急性牙周炎(acute periodontitis,AP)和慢性牙周炎(chronic periodontitis,CP)之间的明确因果关联.方法:使用与9种CTS有关的全基因组关联研究数据(genome-wide association studies,GWAS)和AP、CP人群GWAS数据进行双向MR分析及多变量MR分析.通过利用逆方差加权(inverse variance weighting,IVW)作为主要方法系统性地探讨9种CTS与AP、CP之间的双向因果关系.此外,通过多变量MR分析,确定9种CTS对AP、CP因果作用的独立因果效应.结果:正向MR分析中,IVW结果显示半胱氨酸蛋白酶CTSO与AP之间存在明显的负相关[OR(95%CI)=0.478(0.253~0.901),P=0.022];反向MR分析结果显示,AP与半胱氨酸蛋白酶CTSF之间存在明显的负相关[OR(95%CI)=0.987(0.979~0.996),P=0.005];CP与丝氨酸蛋白酶CTSG[OR(95%CI)=1.027(1.000~1.055),P=0.046]及CTSS[OR(95%CI)=1.035(1.002~1.069),P=0.037]之间存在明显的正相关.多变量MR分析结果显示,CTSG与AP之间呈负相关[OR(95%CI)=0.493(0.282~0.861),P=0.013];CTSS与AP之间呈正相关[OR(95%CI)=1.449(1.012~2.074),P=0.042].结论:CTS在AP、CP发展中具有调节作用,这一发现为CTS与AP、CP之间的关系提供新思路,并且为其预防和治疗提供新的线索.
Objective:To explore the clear causal association between 9 types of cathepsin(CTS)and acute periodontitis(AP)and chronic periodontitis(CP)using Mendelian randomization(MR).Methods:Genome-wide association studies(GWAS)data related to 9 CTSs and GWAS data of AP and CP populations were used for two-way MR analysis and multivariate MR analysis.The two-way causal relationship between the 9 CTSs and AP and CP was systematically explored by using inverse variance weighting(IVW)as the main method,and the independent causal effect CTS of the 9 CTSs on AP and CP were deter-mined by multivariate MR analysis.Results:In the forward MR analysis,the IVW results showed that there was a significant neg-ative correlation between CTSO and AP[OR(95%CI)=0.478(0.253-0.901),P=0.022].The results of reverse MR analysis showed that there was a significant negative correlation between AP and CTSF[OR(95%CI)=0.987(0.979-0.996),P=0.005].There was a significant positive correlation between CP and CTSG[OR(95%CI)=1.027(1.000-1.055),P=0.046]and CTSS[OR(95%CI)=1.035(1.002-1.069),P=0.037].The results of multivariate MR analysis showed that there was a negative corre-lation between CTSG and AP[OR(95%CI)=0.493(0.282-0.861),P=0.013].There was a positive correlation between CTSS and AP[OR(95%CI)=1.449(1.012-2.074),P=0.042].Conclusion:CTS plays a regulatory role in the development of AP and CP,which provides a new idea for the relationship between CTS and AP and CP,and provides new clues for its pre-vention and treatment.
