MTHFS基因多态性及单倍型与老年高血压易感性的关联研究OA
Association between MTHFS gene polymorphisms and haplotypes and susceptibility to hypertension in the elderly
目的:高血压是一种全球流行的慢性疾病,在老年人群中很常见.虽然目前已鉴定出大量与高血压相关的单核苷酸多态性(single nucleotide polymorphisms,SNPs),但仍需进一步探索.本研究探究了MTHFS基因多态性与老年高血压易感性之间的关系.方法:招募583例高血压患者和421例健康对照者.使 用 Agena MassARRAY 平 台 对 5 个 MTHFS SNPs(rs685487、rs622506、rs6495446、rs2562744 和rs2586179)进行基因分型.通过PLINK软件进行Logistic回归分析,以评估SNP与高血压的相关性.使用Sangerbox软件生成了具有明显关联结果的森林图.使用Haploview和SNPStats软件进行单倍型分析.多因素降维方法评估SNP-SNP相互作用.结果:本研究发现rs2562744 A等位基因与高血压风险增加明显相关.rs2562744的CA或AA基因型携带者患高血压的风险明显高于CC基因型.rs2562744的CA基因型与70岁以上人群高血压风险增加明显相关.在男性中,rs2562744也与高血压风险增加明显相关.由rs685487和rs622506形成的AC单倍型与高血压风险明显相关.rs2562744和rs2586179之间的相互作用可能对预测高血压风险很重要.结论:MTHFS rs2562744多态性与老年人高血压风险增加有关.这一发现为探索MTHFS基因在高血压发病机制中的潜在作用提供了新的线索,并可能为未来针对老年高血压的个性化的预防和治疗策略提供参考.
Objective:To be a globally prevalent chronic disease,hypertension is prevalent in the elderly population.Although a large number of single nucleotide polymorphisms(SNPs)associated with hypertension have been identified,further exploration is needed.This study investigates the association between MTHFS gene polymorphisms and hypertension susceptibility in the el-derly.Methods:We recruited 583 hypertensive patients and 421 healthy controls.Five MTHFS SNPs(rs685487,rs622506,rs6495446,rs2562744,and rs2586179)were genotyped using the Agena MassARRAY platform.PLINK software was used for Logistic regression analysis to evaluate association between SNPs and hypertension risk.The forest plot of significant association results was generated using Sangerbox software.Haplotype analysis was performed using Haploview and SNPStats software.The multifactor dimensionality reduction method assessed SNP-SNP interactions.Results:This study found that the rs2562744 A al-lele was significantly linked to increased hypertension risk.Carriers of CA or AA genotypes of rs2562744 had a significantly in-creased risk of hypertension than CC genotypes.The CA genotype of rs2562744 was significantly associated with an increased risk of hypertension in individuals over 70 years old.In males,the rs2562744 also showed a significant association with increased hyper-tension risk.The AC haplotype,formed by rs685487 and rs622506,was significantly linked to hypertension risk.The interaction between rs2562744 and rs2586179 may be important for predicting hypertension risk.Conclusion:The MTHFS rs2562744 poly-morphism is associated with an increased risk of hypertension in the elderly.These findings provide new clues for exploring the po-tential role of the MTHFS gene in the pathogenesis of hypertension and may provide a reference for personalized prevention and treatment strategies for elderly hypertension in the future.
