[目的]系统探究6-苄氨基嘌呤(6-BA)与生长素(IBA)浓度配比对试管苗生长特性、扩繁效率及生理指标的调控效应,优化吴起楸子的离体快繁体系.[方法]试验设置5个6-BA浓度梯度(0.4、0.5、0.6、0.7、0.8 mg/L)和5个IBA浓度梯度(0.5、0.6、0.7、0.8、0.9 mg/L),采用双因素完全随机设计,共10个处理组合,并以无激素MS培养基为对照(CK).培养30 d后,测定扩繁系数、株高、茎粗、节间长度和叶绿素含量等指标.[结果]6-BA与IBA配比显著影响试管苗生长,其中T5-2(6-BA 0.7 mg/L+IBA 0.6 mg/L)的扩繁系数最高(14.125),而T5-1(6-BA 0.5 mg/L+IBA 0.9 mg/L)在茎粗和叶绿素含量上表现最优.[结论]主成分分析表明,6-BA/IBA比值在1.0~1.2时(如1.17)能协同提升增殖效率与生理指标,有效解决徒长问题(节间长度<5.0 mm).研究成功建立了高效低耗的快繁技术体系,为吴起楸楸子砧木的规模化生产提供了理论依据.

[Objective]To optimize the in vitro rapid propagation system for Malus prunifolia fromWuqi,this study sys-tematically investigated the regulatory effects of 6-Benzylaminopurine(6-BA)and auxin(IBA)concentration ratios in MS medium on the growth characteristics,propagation efficiency,and physiological indicators of tube plantlets.[Methods]A two-factor completely randomized design was employed,with five concentrations of 6-BA(0.4,0.5,0.6,0.7,0.8 mg/L)and five concentrations of IBA(0.5,0.6,0.7,0.8,0.9 mg/L),forming 10 treatment combinations(T1-1 to T5-2).A hormone-free MS medium served as the control(CK).After 30 days of culture,parameters such as propagation coefficient,plant height,stem diameter,internode length,and chlorophyll content were measured.[Results]The ratio of 6-BA to IBA significantly influenced the growth of tube plantlets.The combination of 0.7 mg/L 6-BA and 0.6 mg/L IBA(T5-2)yielded the highest propagation coefficient(14.125),while 0.5 mg/L 6-BA and 0.9 mg/L IBA(T5-1)resulted in the best per-formance in stem diameter and chlorophyll content.[Conclusion]Principal component analysis indicated that a 6-BA/IBA ra-tio between 1.0 and 1.2(e.g.,1.17)synergistically enhanced proliferation efficiency and physiological indicators,effectively addressing the issue of excessive elongation(internode length<5.0 mm).This study successfully established a high-efficiency,low-cost rapid propagation system,providing a theoretical basis for the large-scale production of Malus prunifolia rootstocks.
