[目的]优化花牛苹果砧木试管苗的脱毒技术体系,以获得高质量的脱毒材料.[方法]以花牛苹果砧木G935、青砧1号、怀来海棠、新疆野苹果、平邑甜茶、吴起楸子、山定子7种组培苗为试验材料,系统探讨热处理方式与处理时间对脱毒效果的影响.[结果]热处理方式是影响试管苗的存活率和脱毒效率的关键因素,其中以38℃光照培养10 h+32℃暗培养8 h变温处理效果最佳,植株成活率达72%~90%.在相同热处理方式下,接种部位(茎尖或侧芽)对存活率无显著影响.在最佳变温处理(38℃光照培养10 h+32℃暗培养8 h)基础上,设置不同处理时间(15、20、25、30、35、40、45 d)对花牛苹果砧木试管苗进行培养,并结合2次剥茎尖培养发现,处理35 d的脱毒效果最好,茎尖的脱毒率达到了75%~100%;茎尖脱毒效果显著优于侧芽(38%),前者为后者的2倍以上.[结论]研究确立了以38℃光照培养10 h+32℃暗培养8 h处理35 d并结合2次茎尖培养为核心的高效脱毒方案,并获得7种砧木脱毒试管苗材料.

[Objective]This study aimed to optimize the virus-free technology system for tissue-cultured rootstock seed-lings of Huaniu apples to obtain high-quality virus-free materials.[Methods]Tissue-cultured seedlings of seven Huaniu apple rootstocks,namely G935,Qingzhen 1,Huail Begonia,Xinjiang wild apple,Pingyitiancha,Wuqiqiuzi and Shandingzi,were used as experimental materials.The effects of heat treatment methods and duration on virus elimination efficiency were sys-tematically investigated.[Results]The results showed that the heat treatment method was a key factor affecting the survival rate and virus elimination efficiency of the tissue-cultured seedlings.A variable temperature treatment of 38℃with light for 10 hours followed by 32℃in darkness for 8 hours proved to be the most effective,achieving plant survival rates between 72%and 90%.Under the same heat treatment method,the inoculation site,whether shoot tip or lateral bud,had no significant im-pact on the survival rate.Based on this optimal variable temperature treatment,different treatment durations of 15,20,25,30,35,40 and 45 days were tested in combination with a secondary shoot tip excision and culture.The treatment duration of 35 days achieved the best virus elimination effect,with a virus-free rate of 75%to 100%for shoot tips.The virus elimination efficiency of shoot tips was significantly higher than that of lateral buds,which was only 38%,making the former more than twice as effective as the latter.[Conclusion]This study established an efficient virus-free protocol using a 35-day variable temperature treatment of 38℃with light for 10 hours followed by 32℃in darkness for 8 hours,combined with secondary shoot tip culture.Virus-free tissue-cultured seedling materials for all 7 rootstocks were successfully obtained.
