基于质谱技术的石决明药材及其制品鉴别研究OA
Identification Study on Haliotidis Concha Based on Mass Spectrometry
目的:建立石决明药材、饮片、标准汤剂及配方颗粒的质谱鉴别方法,为石决明的鉴别和质量评价提供参考依据.方法:以碳酸氢铵溶液对石决明的蛋白多肽类成分进行提取,并结合HLB固相萃取小柱对提取液进行除杂和富集目标成分.选择质荷比(m/z)517.27(2+)→432.26、m/z 517.27(2+)→618.36、m/z 540.30(2+)→333.19、m/z 540.30(2+)→819.44作为检测离子对,以Waters BEH C18(100 mm×2.1 mm,1.7 μm)为色谱柱,以乙腈-0.1%甲酸水溶液为流动相,梯度洗脱,流速为0.3 mL/min,柱温为30℃,进样量为2 μL.结果:建立的质谱鉴别方法能有效检测皱纹盘鲍药材、饮片、标准汤剂、配方颗粒中的皱纹盘鲍特征离子对,而不同基原、伪品无检出,专属性较强.结论:本研究建立的皱纹盘鲍质谱鉴别方法,方法简单、高效,可为其质量标准研究提供参考.
Objective:To establish a mass spectrometry(MS)identification method for the crude drugs,decoction pieces,standard decoctions and formula granules of Haliotidis Concha,so as to provide a reference for the identification and quality evaluation of Haliotidis Concha.Methods:Ammonium bicarbonate solution was used to extract the proteins and peptides from Haliotidis Concha;the HLB solid-phase extraction column was adopted for impurity removal and enrichment of the target components in the extract.The selected detection ions were m/z 517.27(2⁺)→432.26,m/z 517.27(2⁺)→618.36,m/z 540.30(2⁺)→333.19 and m/z 540.30(2⁺)→819.44.Separation was performed on a Waters BEH C₁₈column(100 mm×2.1 mm,1.7 μm)with acetonitrile and 0.1%formic acid solution as the mobile phase under gradient elution conditions.The flow rate was set at 0.3 mL/min,the column temperature was maintained at 30℃,and the injection volume was 2 μL.Results:The established MS identification method could effectively detect the characteristic ions of Haliotis discus in its crude drugs,decoction pieces,standard decoctions and formula granules.No characteristic ions were detected in samples from other origins and counterfeit products,indicating the strong specificity of this method.Conclusion:The established MS identification method for Haliotis discus is simple and feasible with good applicability,which can provide a scientific reference for the quality research of Haliotidis Concha.
