Chemical Constituents of Helicteres angustifolia
目的:研究梧桐科山芝麻属植物山芝麻Helicteres angustifolia L.的化学成分.方法:采用硅胶、ODS、Sephadex LH-20 葡聚糖凝胶、制备HPLC等色谱技术对山芝麻根甲醇提取物的乙酸乙酯部位进行分离纯化,并利用现代波谱技术鉴定化合物结构.结果:从山芝麻中分离得到 32 个化合物,分别鉴定为:(2R,8S,9R,10R,13R,14S,16R,17R)-17-[(R,E)-2,6-dihydroxy-6-methyl-3-oxohept-4-en-2-yl]-2,16-dihydroxy-4,4,9,13,14-pentamethyl-1,4,7,8,9,10,12,13,14,15,16,17-dodecahydro-3H-cyclopenta[a]phenanthrene-3,11(2H)-dione(1)、葫芦素I(2)、葫芦素 E(3)、葫芦素 J(4)、hexanorcucurbitacin I(5)、3-O-咖啡酰基齐墩果酸(6)、3β-acetoxy-27-(benzoyloxy)olean-12-en-28-oic acid methyl ester(7)、3β-acetoxy-27-trans-caffeoyloxyolean-12-en-28-oic acid methyl ester(8)、3β-(p-hydroxy-trans-cinnamoyloxy)olean-12-en-28-oic acid(9)、3β-hydraxyolean-12-en-27-benzoyloxy-28-oate(10)、3β-hydroxyoleana-11,13(18)-dien-28-oic acid(11)、3β-acetoxy-27-[(4-hydroxybenzoyl)oxy]olean-12-en-28-oic acid methyl ester(12)、白桦脂酸(13)、圆齿火棘酸(14)、山芝麻酸甲酯(15)、6,7,9α-trihydroxy-3,8,11α-trimethylcyclohexo-[d,e]-coumarin(16)、6,7-dihydroxy-3,8,11-trimethylcyclohexo-[d,e]-coumarin(17)、mansorin I(18)、mansonone F(19)、mansonone E(20)、mansonone H(21)、mansonone M(22)、丁香脂素(23)、二氢去氢二愈创木基醇(24)、buddlenol E(25)、对羟基苯甲酸(26)、ferulyl aldehyde(27)、methyl(E)-octadec-9-enoate(28)、棕榈酸(29)、β-谷甾醇(30)、豆甾醇(31)、hibiscone C(32).结论:其中,化合物 1 为新构型,化合物 6、8、18、25~29、32 为首次从该植物中分离得到.
Objective:To study the chemical constituents of Helicteres angustifolia.Methods:The ethyl acetate fraction of methanol extract from the root of Helicteres angustifolia was isolated and purified by using chromatographic techniques such as silica gel,ODS,Sephadex LH-20 glucan gel,and preparative HPLC,and the compound structures were identified by modern spectroscopy.Results:Thir-ty-two compounds were isolated from Helicteres angustifolia and identified as(2R,8S,9R,10R,13R,14S,16R,17R)-17-[(R,E)-2,6-dihydroxy-6-methyl-3-oxohept-4-en-2-yl]-2,16-dihydroxy-4,4,9,13,14-pentamethyl-1,4,7,8,9,10,12,13,14,15,16,17-dodeca-hydro-3H-cyclopenta[a]phenanthrene-3,11(2H)-dione(1),cucurbitacin I(2),cucurbitacin E(3),cucurbitacin J(4),hexanorcucurb-itacin I(5),oleanolic acid 3β-caffeate(6),3β-acetoxy-27-(benzoyloxy)olean-12-en-28-oic acid methyl ester(7),3β-acetoxy-27-trans-caffeoyloxyolean-12-en-28-oic acid methyl ester(8),3β-(p-hydroxy-trans-cinnamoyloxy)olean-12-en-28-oic acid(9),3β-hydraxyolean-12-en-27-benzoyloxy-28-oate(10),3β-hydroxyoleana-11,13(18)-dien-28-oic acid(11),3β-acetoxy-27-[(4-hydroxybenzoyl)oxy]olean-12-en-28-oic acid methyl ester(12),betulic acid(13),pyracrenic acid(14),methyl helicterate(15),6,7,9α-trihydroxy-3,8,11α-trimethylcyclohexo-[d,e]-coumarin(16),6,7-dihydroxy-3,8,11-trimethylcyclohexo-[d,e]-coumarin(17),mansorin I(18),man-sonone F(19),mansonone E(20),mansonone H(21),mansonone M(22),syringaresinol(23),dihydrodehydroconiferyl alcohol(24),buddlenol E(25),4-hydroxybenzoic acid(26),ferulyl aldehyde(27),methyl(E)-octadec-9-enoate(28),palmitic acid(29),β-sitoster-ol(30),stigmasterol(31),hibiscone C(32),respectively.Conclusion:Among them,compound 1 is a new configuration,and compounds 6,8,18,25 to 29 and 32 are isolated from Helicteres angustifolia for the first time.
