特异性PCR法鉴别白僵菌及其混伪品OA

目的:建立一种能够快速鉴别僵蚕及其混伪品的特异性聚合酶链式反应(PCR)方法.方法:将测序得到的感染僵蚕的菌种白僵菌Beauveria bassiana ITS序列与近源物种基因序列进行比对,根据变异位点较多的片段设计特异性引物TF2/TR2,并对PCR反应条件进行优化和验证.结果:当退火温度为 64℃,循环 30 次时,14 批僵蚕、含僵蚕的药材粉末及白僵菌标准样品均可在 258 bp处扩增得到单一清晰的目标条带,而其余 27 批混伪品均无此条带.结论:该研究所建立的特异性PCR方法准确地实现了僵蚕药材及其混伪品的真伪鉴定,该方法专属性良好,操作方便快捷,适用于僵蚕药材的质量控制,为解决僵蚕药材的品质稳定性和安全性问题提供了技术保障.

Objective:To establish a specific polymerase chain reaction(PCR)method for the rapid identification of Bombyx Ba-tryticatus and its adulterants.Methods:The ITS sequence of Beauveria bassiana,a fungal species associated with Bombyx Batryticatus,was compared with the corresponding gene sequences of related species.The specific primer TF2/TR2 was designed based on the frag-ments with a high number variation sites,and the PCR reaction conditions were optimized and validated.Results:At an annealing tem-perature of 64℃and with 30 cycles,all 14 batches of Bombyx Batryticatus,the powdered medicinal materials containing Bombyx Ba-tryticatus,and standard samples of Beauveria bassiana yielded a single clear target band at 258 bp.In contrast,none of the 27 batches of adulterants displayed this band.Conclusion:The specific PCR method established in this research institute accurately realizes the au-thenticity identification of Bombyx Batryticatus and its adulterants.This method has good specificity,is convenient and fast to operate,and is suitable for the quality control of Bombyx Batryticatus,providing technical support for addressing the issues of the stability and safety of Bombyx Batryticatus medicinal material.
