Research progress on the mechanism of Astragali Radix in treating Alzheimer's Disease

阿尔茨海默病(AD)是一种严重的神经退行性疾病,其发病机制复杂,临床缺乏根治手段.黄芪作为传统中药,在AD防治中展现出多靶点干预的潜力.本文通过系统综述发现,黄芪及其活性成分(如黄芪甲苷、黄芪多糖)不仅可通过调控Aβ稳态、抑制神经细胞凋亡、保护血脑屏障等途径发挥作用,新近研究还证实其能直接或间接抑制Tau蛋白过度磷酸化,干预AD核心病理.在机制上,黄芪主要通过调控细胞凋亡信号通路、NF-κB/Nrf2介导的神经炎症与氧化应激核心通路、PPARγ/GSK-3β等Tau相关通路以及MMPs系统,发挥多靶点协同治疗效应.本文系统梳理了黄芪治疗AD的分子网络,为后续深入研究与临床转化提供了重要理论参考;同时指出,当前研究在Tau蛋白直接机制、黄酮类成分探索及临床转化方面仍存不足,是未来重点研究方向.

Alzheimer's disease(AD)is a severe neurodegenerative disorder with complex pathogenesis and no definitive clinical cure.As a traditional Chinese medicine,Astragali Radix has demonstrated potential for multi-target intervention in AD prevention and treatment.Through systematic review,this study reveals that Astragali Radix and its active components(such as astragaloside IV and polysaccharides)exert effects not only by regulating Aβ homeostasis,inhibiting neuronal apoptosis,and protecting the blood-brain barrier but also by directly or indirectly suppressing Tau protein hyperphosphorylation,addressing the core pathology of AD.Mecha-nistically,Astragali Radix primarily exerts its multi-target synergistic therapeutic effects by modulating the apoptosis signaling path-way,the NF-κB/Nrf2-mediated neuroinflammation and oxidative stress core pathway,Tau-related pathways such as PPARγ/GSK-3β,and the MMPs system.This study systematically maps the molecular network of Astragali Radix in treating AD,providing important theoretical references for further research and clinical translation.However,it also highlights current gaps in understanding the direct mechanisms of Tau protein,exploration of flavonoid components,and clinical translation,which remain key future research directions.
