基于LC-MS/MS技术的四妙汤水煎液及入血成分分析OA
Analysis of Simiao Decoction and Absorbed Compounds in the Blood Based on LC-MS/MS
目的:利用血清药物化学方法对四妙汤水煎液和入血成分进行分析.方法:采用液相色谱-串联质谱(LC-MS/MS)技术检测水煎液及大鼠血清中的原型成分及代谢成分,并对其代谢途径进行分析.结果:从四妙汤水煎液中得到核心对比成分114 个;从空白血清中得到核心对比成分148 个;从四妙汤含药血清中得到核心对比成分158 个.结论:初步确定了 7α-羟胆固醇、牛磺胆酸、甘氨胆酸、3α,7α-二羟-5β-胆烷酸等成分可能为四妙汤入血的药效物质基础,且胆汁酸代谢可能为四妙汤入血的主要代谢途径.
Objective:To analyze the decoction and the absorbed compounds in the blood of Simiao Decoction based on the serum drug chemistry method.Methods:LC-MS/MS was used to detect the prototype and metabolic components in the decoction and rat ser-um,and their metabolic pathways were analyzed.Results:114 core comparative components were obtained from Simiao Decoction;148 core comparative components were obtained from blank serum;The serum containing Simiao Decoction obtained 158 core comparative components.Conclusion:It has been preliminarily determined that components such as 7α-hydroxycholesterol,taurocholic acid,glyco-cholic acid,and 3α,7α-dihydroxy-5β-cholic acid may be the material basis for the pharmacological effects of Simiao Decoction when it enters the bloodstream,and bile acid metabolism may be the main metabolic pathway for Simiao Decoction entering the bloodstream.
