艾灸对克罗恩病大鼠结肠H3K27乙酰化的调节作用研究OA
Study on the regulation effect of moxibustion on colonic H3K27 acetylation in rats with Crohn disease
目的:观察艾灸对克罗恩病(CD)大鼠的干预效应,探讨干预效应与结肠组蛋白H3第27位赖氨酸(H3K27)乙酰化(H3K27ac)作用水平变化的关系.方法:根据随机数字表法从36只Sprague-Dawley大鼠中随机挑选10只作为对照组,其余26只大鼠采用2,4,6三硝基苯磺酸(TNBS)灌肠制备大鼠CD模型.将造模成功的大鼠根据随机数字表法分为模型组、隔药灸组和柳氮磺吡啶组,每组8只.隔药灸组和柳氮磺吡啶组分别采用隔药灸气海、双侧天枢和柳氮磺吡啶灌胃干预.运用苏木素-伊红染色观察CD大鼠结肠组织病理变化.酶联免疫吸附法检测血清炎症因子[肿瘤坏死因子(TNF)-α、白细胞介素(IL)-1β和IL-6]水平.免疫印迹法及免疫组化检测结肠H3K27ac蛋白表达.实时荧光定量逆转录聚合酶链反应和免疫荧光检测赖氨酸乙酰转移酶2B(KAT2B)、一般控制非阻遏蛋白5(GCN5)和组蛋白去乙酰化酶1(HDAC1)、组蛋白去乙酰化酶3(HDAC3)mRNA和蛋白表达情况.结果:与对照组比较,模型组结肠组织病理学评分、血清炎症因子TNF-α、IL-1β、IL-6蛋白含量和结肠GCN5、HDAC1、HDAC3 mRNA和蛋白表达均显著升高(P<0.01或P<0.05),H3K27ac蛋白、KAT2B mRNA和蛋白表达均显著降低(P<0.01或P<0.05).与模型组比较,隔药灸组大鼠结肠黏膜损伤明显改善,结肠组织病理学评分、TNF-α、IL-1β、IL-6的蛋白含量和GCN5、HDAC1、HDAC3 mRNA和蛋白表达均显著降低(P<0.01或P<0.05),H3K27ac蛋白和KAT2B mRNA和蛋白表达均显著升高(P<0.01或P<0.05).柳氮磺吡啶组与隔药灸组比较,上述指标差异无统计学意义.结论:艾灸能够减轻CD大鼠结肠炎症反应;可能与上调结肠组织KAT2B表达,同时下调GCN5、HDAC1和HDAC3的表达,从而增加H3K27ac水平有关.
Objective:To investigate the regulatory effect of moxibustion on rats with Crohn disease(CD)and explore the relationship between the effect and alterations in colonic histone H3 lysine 27 acetylation(H3K27ac). Methods:Ten out of 36 Sprague-Dawley rats were randomly selected as the control group using a random number table method.The remaining 26 rats received an enema of 2,4,6-trinitrobenzene sulfonic acid to establish a CD model.Successfully modeled rats were then randomly divided into a model group,an herb-insulated moxibustion group,and a sulfasalazine group using the random number table method,with 8 rats in each group.The herb-insulated moxibustion group received herb-insulated moxibustion at Qihai(CV6)and bilateral Tianshu(ST25),and the sulfasalazine group received sulfasalazine via gavage.Histopathological changes in the colonic tissues were assessed using hematoxylin-eosin staining.The serum levels of inflammatory factors[tumor necrosis factor(TNF)-α,interleukin(IL)-1β,and IL-6]were measured using the enzyme-linked immunosorbent assay.The protein expression of colonic H3K27ac was detected using Western blotting and immunohistochemistry.The mRNA and protein expression levels of K-acetyltransferase 2B(KAT2B),general control non-repressed protein 5(GCN5),histone deacetylase 1(HDAC1),and histone deacetylase 3(HDAC3)in the colon were determined using real-time quantitative reverse transcription polymerase chain reaction and immunofluorescence,respectively. Results:Compared to the control group,the histopathological score,serum levels of TNF-α,IL-1β,and IL-6,and the mRNA and protein expression of GCN5,HDAC1,and HDAC3 in the colon were significantly increased in the model group(P<0.01 or P<0.05),whereas the protein expression of H3K27ac and the mRNA and protein expression of KAT2B were significantly decreased in the model group(P<0.01 or P<0.05).In contrast,compared to the model group,the histopathological score,serum levels of TNF-α,IL-1β,and IL-6,and the mRNA and protein expression of GCN5,HDAC1,and HDAC3 in the colon were significantly decreased in the herb-insulated moxibustion group(P<0.01 or P<0.05),whereas the protein expression of H3K27ac and the mRNA and protein expression of KAT2B were significantly increased in the model group(P<0.01 or P<0.05).No significant differences in the aforementioned indicators were observed between the sulfasalazine group and the herb-insulated moxibustion group. Conclusion:Moxibustion alleviates colonic inflammation in CD rats,potentially by up-regulating the expression of KAT2B,down-regulating the expression of GCN5,HDAC1,and HDAC3,and consequently increasing the H3K27ac level.
