基于响应面法优化沙棘制汁的酶解工艺OA

Optimization of enzymatic process for sea buckthorn juice production based on response surface methodology

目的:优化沙棘汁的酶法提取工艺,以提升其作为中药口服液、合剂等液体制剂原料的提取效率与质量.方法:以沙棘为原料,采用果胶酶与纤维素酶进行复配酶解.在单因素实验基础上,选取复合酶比例、酶添加量、酶解温度、酶解时间和pH值为关键因素,运用响应面法进行工艺优化,并以沙棘出汁率作为核心评价指标.结果:确定了最佳酶解工艺参数为:纤维素酶与果胶酶比例1﹕2,酶添加量1.5%,酶解温度54℃,酶解时间107 min,酶解pH值5.5.在此条件下,沙棘出汁率最高,达到(84.63±1.47)%.结论:本研究建立的提取工艺稳定、高效,可为沙棘汁作为中药制剂原料的标准化、工业化生产提供技术依据,有助于推动沙棘在中药领域的进一步开发利用.

Objective:To optimize the enzymatic extraction process of sea buckthorn juice to enhance its extraction efficiency and quality as a raw material for liquid preparations of Chinese medicine,such as oral liquids and mixtures.Methods:Using sea buckthorn as the raw material,combined enzymatic hydrolysis was performed with pectinase and cellulase.Key factors,including the compound enzyme ratio,enzyme dosage,enzymatic hydrolysis temperature,time,and pH,were investigated.Based on single-factor experiments,the extraction process was optimized using response surface methodology,with the juice yield of sea buckthorn as the primary evaluation index.Results:The optimal enzymatic hydrolysis parameters were determined as follows:cellulase to pectinase ratio of 1:2,enzyme dosage of 1.5%,enzymatic hydrolysis temperature of 54 ℃,time of 107 min,and pH value of 5.5.Under these conditions,the highest sea buckthorn juice yield reached(84.63±1.47)%.Conclusion:The established extraction process is stable and efficient,providing a technical basis for the standardized and industrial production of sea buckthorn juice as a raw material for Chinese medicinal preparations.This study supports the further development and utilization of sea buckthorn in the field of Chinese medicine.
