应用环介导等温扩增技术鉴别毛慈菇与冰球子OA
Identification of Cremastra appendiculata and Pleione bulbocodioides Using Loop-Mediated Isothermal Amplification Technology
目的:建立毛慈菇与冰球子两种中药材相互快速鉴别的特异性环介导等温扩增技术(LAMP)体系.方法:选择叶绿体片段中rbcL编码区片段作为检测靶标,设计可用于甄别毛慈菇与冰球子的特异性引物,建立LAMP鉴别方法,并对其进行特异性、灵敏度、适用性等方法学指标进行考察.结果:所建立的LAMP 快速检测方法在反应温度为65℃,反应时间为 60 min的条件下即可有效识别毛慈菇与冰球子这两种易混中药材,其检测灵敏度可达到 500 ag/μL,且反应结果易于观察.结论:该研究建立的中药材毛慈菇与冰球子LAMP快速鉴别方法具有灵敏度高、特异性好、快速实用、易于推广等特点,适用于毛慈菇和冰球子两种中药材的质量监控.
Objective:To establish a specific loop-mediated isothermal amplification(LAMP)method system for the rapid identifi-cation of two Chinese herbal medicines,Cremastra appendiculata and Pleione bulbocodioides.Methods:The rbcL coding region fragment in the chloroplast fragment was selected as the detection target.Specific primers for the identification of Cremastra appendiculata and Pleione bulbocodioides were designed.The LAMP identification method was established,and its methodological indicators such as speci-ficity,sensitivity and applicability were investigated.Results:The established LAMP rapid detection method under the reaction tempera-ture of 65℃and reaction time of 60 min could effectively distinguish the two easily-confused Chinese medicinal materials Cremastra appendiculata and Pleione bulbocodioides.The detection sensitivity could reach 500 ag/μL,and the reaction results were easy to ob-serve.Conclusion:The LAMP rapid identification method for the Chinese medicinal materials Cremastra appendiculata and Pleione bul-bocodioides established in this study has the characteristics of high sensitivity,good specificity,rapidity and practicality,and easy promo-tion.It is suitable for the quality monitoring of two Chinese medicinal materials Cremastra appendiculata and Pleione bulbocodioides.
