基于多指标成分定量联合化学计量学及熵权TOPSIS模型评价不同产地天葵子的质量OA
Quality Evaluation of Semiaquilegiae Radix from Different Origins Based on Multi-Component Quantification Combined with Chemometrics and Entropy Weight-TOPSIS Model
目的 建立天葵子多指标成分定量检测方法,并联合化学计量学及熵权TOPSIS模型评价不同产地天葵子的质量.方法 采用Develosil ODS-MG-5 C18 色谱柱,以乙腈-0.1%磷酸为流动相,梯度洗脱,切换波长(254、210 nm),以高效液相色谱法(HPLC)同时测定 18 批天葵子中 11 个成分(东方唐松草苷、紫草氰苷、蝙蝠葛氰苷、格列风内酯、阿魏酸、原儿茶酸、木兰花碱、唐松草酚定、小檗红碱、胡萝卜苷、β-谷甾醇)的含量,并检测其醇溶性浸出物、总灰分和酸不溶性灰分的含量.采用聚类分析(CA)、主成分分析(PCA)、正交偏最小二乘法判别分析(OPLS-DA)和熵权TOPSIS模型综合评价不同产地天葵子的质量.结果 18 批天葵子中东方唐松草苷、紫草氰苷、蝙蝠葛氰苷、格列风内酯、阿魏酸、原儿茶酸、木兰花碱、唐松草酚定、小檗红碱、胡萝卜苷、β-谷甾醇的含量分别为 0.692~1.185、1.824~3.132、0.531~1.078、0.223~0.544、0.202~0.392、0.987~1.948、0.825~2.212、0.380~0.812、0.112~0.328、0.050~0.104、0.349~0.558 mg·g-1;醇溶性浸出物、总灰分和酸不溶性灰分含量分别为 12.495%~20.139%、3.125%~6.477%、1.120%~3.543%.CA、PCA结果显示,18 批天葵子可分为 3 类,分别为S1~S7、S8~S12 和S13~S18;OPLS-DA筛选出VIP值>1影响天葵子质量差异的标志物 5 个,分别为紫草氰苷、木兰花碱、原儿茶酸、蝙蝠葛氰苷、格列风内酯.熵权TOPSIS模型结果显示,18 批样品的最优解欧氏贴近度均值范围为 0.130 7~0.772 7,不同产地天葵子存在质量差异.结论 该研究建立的多指标含量测定方法准确、可靠,联合化学计量学及熵权TOPSIS模型能够对不同产地天葵子的药材质量进行评价,可为天葵子药材质量控制、综合评价提供更多参考.
Objective To establish a multi-component quantitative detection method for Semiaquilegiae Radix and evaluate the quality of samples from different origins by combining chemometrics and an entropy weight-TOPSIS model.Methods Using a Develosil ODS-MG-5 C18 column with acetonitrile-0.1%phosphoric acid as the mobile phase under gradient elution and wavelength switching(254,210 nm),an HPLC method was developed to simultaneously determine the contents of 11 components(thalictoside,lithospermoside,menisdaurin,griffonilide,ferulic acid,protocatechuic acid,magnoflorine,thalifendine,berberrubine,daucosterol,and β-sitosterol)in 18 batches of Semiaquilegiae Radix.The contents of alcohol-soluble extractives,total ash,and acid-insoluble ash were also measured.Cluster analysis(CA),principal component analysis(PCA),orthogonal partial least squares-discriminant analysis(OPLS-DA),and an entropy weight-TOPSIS model were employed to comprehensively evaluate the quality of Semiaquilegiae Radix from different origins.Results The contents of the 11 components in the 18 batches were as follows:thalictoside 0.692-1.185 mg·g-1,lithospermoside 1.824-3.132 mg·g-1,menisdaurin 0.531-1.078 mg·g-1,griffonilide 0.223-0.544 mg·g-1,ferulic acid 0.202-0.392 mg·g-1,protocatechuic acid 0.987-1.948 mg·g-1,magnoflorine 0.825-2.212 mg·g-1,thalifendine 0.380-0.812 mg·g-1,berberrubine 0.112-0.328 mg·g-1,daucosterol 0.050-0.104 mg·g-1,and β-sitosterol 0.349-0.558 mg·g-1.The contents of alcohol-soluble extractives,total ash,and acid-insoluble ash were 12.495%~20.139%,3.125%~6.477%,and 1.120%~3.543%,respectively.CA and PCA results indicated that the 18 batches could be classified into three categories:S1-S7,S8-S12,and S13-S18.The OPLS-DA model screened five markers with VIP values>1 that significantly influenced quality variation:lithospermoside,magnoflorine,protocatechuic acid,menisdaurin,and griffonilide.The entropy weight-TOPSIS model results showed that the Euclidean closeness degree to the optimal solution for the 18 samples ranged from 0.130 7 to 0.772 7,indicating quality differences among Semiaquilegiae Radix from different origins.Conclusion The established multi-component quantification method is accurate and reliable.Combined with chemometrics and the entropy weight-TOPSIS model,it can effectively evaluate the quality of Semiaquilegiae Radix from different origins,providing a valuable reference for its quality control and comprehensive assessment.
