目的:研究九味健步饮含药血清对LPS诱导大鼠CP-R092 细胞凋亡及炎症的影响.方法:构建软骨细胞退变模型,设空白对照组、模型组、九味健步饮含药血清组、LY294002(PI3K抑制剂)组、九味健步饮含药血清+LY294002 组.显微镜下观察各组软骨细胞形态变化;CCK-8法检测各组细胞活力;流式细胞术检测各组细胞凋亡率;免疫荧光染色检测Bax蛋白表达;RT-qPCR、Western Blot检测软骨细胞PI3K、AKT、mTOR、Bax、Bcl-2、IL-6、TNF-α mRNA和蛋白表达.结果:与空白对照组比较,模型组细胞数量减少,形态发生变化,细胞活力显著降低,凋亡率显著升高,PI3K、mTOR、Bax、IL-6、TNF-α mRNA和蛋白表达及AKT mRNA和p-AKT蛋白表达显著升高,Bcl-2 mR-NA和蛋白表达显著降低(P<0.05 或 P<0.01).与模型组比较,各给药组细胞数量增多,形态变化不大,细胞活力显著降低,凋亡率显著升高,PI3K、mTOR、Bax、IL-6、TNF-α mRNA和蛋白表达及AKT mRNA和p-AKT蛋白表达显著降低,Bcl-2 mRNA和蛋白表达显著升高(P<0.05).结论:九味健步饮含药血清可抑制PI3K/AKT/mTOR信号通路,减轻炎症,减少软骨细胞凋亡.

Objective:To evaluate the effect of drug-containing serum of Jiuwei Jianbu Decoction on apoptosis and inflammation of LPS-induced rat CP-R092 cells.Methods:The chondrocyte degeneration model was established,the blank control group,model group,Jiuwei Jianbu Decoction drug-containing serum group,LY294002(PI3K inhibitor)group,Jiuwei Jianbu Decoction drug-containing ser-um+LY294002 group were set up.The morphological changes of chondrocytes in each group were observed under the microscope;The cell viability of each group was detected by the CCK-8 method;The apoptosis rate of cells in each group was detected by flow cytometry;Immunofluorescence staining was used to detect the expression of Bax protein;The expressions of PI3K,AKT,mTOR,Bax,Bcl-2,IL-6 and TNF-α mRNA and proteins in chondrocytes were detected by RT-qPCR and Western Blot.Results:Compared with the blank control group,the number of cells in the model group decreased,the morphology changed,the cell viability significantly decreased,the apoptosis rate significantly increased,and the mRNA and protein expressions of PI3K,mTOR,Bax,IL-6,and TNF-α and the AKT mRNA and p-AKT protein significantly increased.The expressions of Bcl-2 mRNA and protein were significantly decreased(P<0.05 or P<0.01).Compared with the model group,the number of cells in each administration group increased,the morphological changes were not signifi-cant,the cell viability increased significantly,the apoptosis rate decreased significantly,the mRNA and protein expressions of PI3K,mTOR,Bax,IL-6,TNF-α and the AKT mRNA and p-AKT protein decreased significantly,and the mRNA and protein expressions of Bcl-2 increased significantly(P<0.05).Conclusion:The drug-containing serum of Jiwei Jianbu Decoction can inhibit the PI3K/AKT/mTOR signaling pathway,alleviate inflammation,and reduce chondrocyte apoptosis.
