目的:探讨益气散瘀解毒方主要作用成分通过调控REGγ/RNF149/BRAF/MEK/ERK信号通路抑制骨肉瘤发生发展的作用机制.方法:对9位骨肉瘤患者的邻近正常组织和骨肉瘤组织样本进行质谱分析及免疫组织化学实验检测,以骨肉瘤细胞143B和 MG63为研究对象,通过shRNA干扰技术和瞬时转染方法进行REGγ蛋白表达调控,采用Western Blot实验和RT-PCR实验进行转染效率验证及RNF149、BRAF、MEK、ERK表达水平检测,采用蛋白质免疫共沉淀实验和体外降解实验验证REGγ和RNF149蛋白的相互作用;采用高效液相色谱法鉴定复方中的黄芪、黄连和重楼颗粒剂中的主要成分,分别处理细胞后通过CCK-8实验、细胞平板克隆实验、Transwell实验、Western Blot实验、RT-PCR实验检测细胞增殖能力及REGγ/RNF149/BRAF/MEK/ERK 信号通路表达水平;通过瞬时转染方法进行REGγ过表达,采用CCK-8实验、细胞平板克隆实验检测黄连素、重楼皂苷Ⅰ及中药复方对两种骨肉瘤细胞的毒性作用,采用 Western Blot实验和RT-PCR实验检测基因表达水平变化;用MEK/ERK通路激活剂C16-PAF单独/联合中药复方处理细胞,用CCK-8法检测细胞存活率.结果:REGγ在骨肉瘤组织中高表达;敲低REGγ可以显著抑制两种骨肉瘤细胞的增殖和侵袭能力,RNF149的mRNA水平无显著变化,蛋白水平均显著升高,BRAF、p-MEK、p-ERK蛋白水平显著降低;REGγ参与调控 RNF149蛋白的泛素和 ATP非依赖的蛋白降解过程;黄连素、重楼皂苷Ⅰ是益气散瘀解毒方中发挥抑癌作用的主要成分,且中药复方较两种有效单体成分更显著抑制了两种骨肉瘤细胞的增殖能力,与敲低REGγ后的蛋白表达水平变化一致;过表达REGγ后黄连素、重楼皂苷Ⅰ及中药复方对两种骨肉瘤细胞的毒性作用被显著抑制,过表达REGγ后中药复方上调RNF149蛋白表达及下调BRAF、p-MEK、p-ERK蛋白表达水平的能力被显著抑制;C16-PAF可以显著抑制黄连素、重楼皂苷Ⅰ及中药复方对两种骨肉瘤细胞的毒性作用.结论:益气散瘀解毒方通过 REGγ/RNF149/BRAF/MEK/ERK 信号通路参与调控骨肉瘤的发生发展,其中黄连素、重楼皂苷Ⅰ是其主要作用成分.

Objective:To explore the mechanism by which the main active components of the Yi-Qi-San-Yu-Jie-Du formula inhibit the occurrence and development of osteosarcoma through regulating the REGγ/RNF149/BRAF/MEK/ERK signa-ling pathway.Methods:Mass spectrometry and immunohistochemical experiments were conducted on samples of adjacent normal tissues and osteosarcoma tissues from 9 osteosarcoma patients.Osteosarcoma cell lines 143B and MG63 were used as research subjects.The expression of REGγ protein was regulated by shRNA interference and transient transfection methods.Western Blot and RT-PCR were used to verify the transfection efficiency and detect the expression levels of RNF149,BRAF,MEK,and ERK.Co-immunoprecipitation experiments and in vitro degradation experiments were per-formed to verify the interaction between REGγ and RNF149 proteins.The main components of Astragalus,Coptis,and Polyphyllin in the compound were identified by high-performance liquid chromatography.After treating the cells with these components,CCK-8,cell plate cloning,Transwell,Western Blot,and RT-PCR were used to detect cell proliferation ability and the expression levels of the REGγ/RNF149/BRAF/MEK/ERK signaling pathway.REGγ overexpression was achieved by transient transfection,and CCK-8 and cell plate cloning experiments were used to detect the cytotoxic effects of berberine,polyphyllin Ⅰ,and the compound on the two osteosarcoma cell lines.Western Blot and RT-PCR were used to detect changes in gene expression levels.C16-PAF,an activator of the MEK/ERK pathway,was used to treat the cells alone or in combination with the compound,and CCK-8 was used to detect cell survival rates.Results:REGγ was highly expressed in osteosarcoma tissues.Knockdown of REGγ significantly inhibited the proliferation and invasion abilities of the two osteosarcoma cell lines.The mRNA level of RNF149 did not change significantly,but the protein level increased significantly.The protein levels of BRAF,p-MEK,and p-ERK decreased significantly.REGγ was involved in regulating the ubiquitin and ATP-independent protein degradation process of RNF149.Berberine and polyphyllin Ⅰ were the main components of the Yi-Qi-San-Yu-Jie-Du formula that exerted anti-cancer effects,and the compound more significantly inhibited the proliferation ability of the two osteosarcoma cell lines than the two effective monomer components,which was consistent with the changes in protein expression levels after REGγ knockdown.Overexpression of REGγ significantly inhibited the cytotoxic effects of berberine,polyphyllin Ⅰ,and the compound on the two osteosarcoma cell lines.Overex-pression of REGγ significantly inhibited the ability of the compound to upregulate RNF149 protein expression and down-regulate BRAF,p-MEK,and p-ERK protein expression levels.C16-PAF significantly inhibited the cytotoxic effects of berberine,polyphyllin Ⅰ,and the compound on the two osteosarcoma cell lines.Conclusion:Yi-Qi-San-Yu-Jie-Du formula participates in the regulation of osteosarcoma occurrence and development through the REGγ/RNF149/BRAF/MEK/ERK signaling pathway,and berberine and polyphyllin Ⅰ are its main active components.
