该研究从"二次打击"理论出发,探讨了病证结合动物模型切实可行的研究思路和方法.病证结合动物模型在中医药研究中具有重要意义,不仅能还原疾病的病理过程,还能体现中医证候特征,为中医药复方及新药研发提供科学依据."二次打击"理论通过初次和二次打击的协同作用,模拟疾病的多阶段性和多因素性,弥补了单一因素刺激诱导病证结合动物模型的不足,能够更真实地反映疾病病理和证候特性.通过分析病证结合动物模型研究现状及"二次打击"致病理论的优势,阐明其在揭示疾病复杂发病机制、发现新靶点、推动个体化医疗及提高实验动物模型的临床外推性等方面的价值.同时在此基础上进一步探讨了该理论在构建符合中医特点的病证结合动物模型中的潜在价值和具体应用方法,包括刺激因素的选择、病理机制的契合性分析以及客观化评价方法的建立.总体而言,"二次打击"理论为中医病证结合动物模型研究提供了新思路和新方法,促进了中西医结合研究的发展,当然在研究过程中一定程度上也面临刺激因素选择和动物伦理等方面的挑战.
This study explores the research ideas and methods for establishing the animal models combining disease with syndrome from the"two-hit"theory.Such animal models hold importance in TCM research as they not only replicate the pathological processes of diseases but also embody the characteristics of TCM syndromes.These models provide a scientific basis for the development of TCM formulas and novel drugs.The"two-hit"theory simulates the multi-stage and multi-factorial nature of diseases through the synergistic effects of primary and secondary hits,compensating for the limitations of single-factor models and authentically reflecting the pathological and syndrome-specific characteristics of diseases.This article analyzes the advantages of the"two-hit"theory and elucidates its value in uncovering the complex mechanisms of disease onset,identifying new therapeutic targets,promoting personalized medicine,and enhancing the clinical translatability of animal models.Additionally,the article discusses the specific applications of this theory in constructing animal models combining disease with syndrome that align with TCM principles.These applications include the selection of stimuli,the analysis of pathophysiological congruence,and the establishment of objective evaluation methods.The"two-hit"theory introduces new research ideas and methods for the animal models combining disease with syndrome in TCM,facilitating the integration of TCM and western medicine.However,it also faces challenges such as the selection of stimuli and ethical considerations.
