Multidimensional Inhibitory Effects of Lycopene on Staphylococcus aureus-Induced Mastitis in Mice

为了探究番茄红素(LYC)对金黄色葡萄球菌(S.aureus)诱导小鼠乳腺炎的保护作用及其机制,本试验选取6~8周龄昆明妊娠母鼠36只,随机均分为6个组:对照组、LYC组、模型组、低剂量组、中剂量组和高剂量组.从母鼠配种中期(即母鼠见栓后第7天)开始,LYC组、低剂量组、中剂量组和高剂量组每天分别灌胃给予50、12.5、25和50 mg/(kg·bw)LYC,对照组和模型组灌胃同体积的玉米油,连续灌胃28 d后,于乳腺导管注射S.aureus悬液(2×108 CFU/mL)建立小鼠乳腺炎模型(对照组和LYC组不做处理).24 h后处死小鼠,观察小鼠乳腺组织解剖学变化;摘取第4对乳腺组织,苏木精-伊红(H.E.)染色观察乳腺组织病理学变化;酶联免疫吸附试验(ELISA)检测小鼠乳腺组织炎症因子[肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、白细胞介素-1β(IL-1β)]浓度;蛋白免疫印迹(WB)检测紧密连接蛋白[闭合小环蛋白1(ZO-1)、闭合蛋白(Occludin)、紧密连接蛋白-3(Claudin-3)]表达.进一步通过微量肉汤法检测LYC对S.aureus的最小抑菌浓度(MIC),绘制S.aureus生长曲线;溶血法检测S.aureus溶血活性;WB检测S.aureus的α-溶血素(Hla)表达.结果显示,与对照组相比,模型组小鼠乳腺组织出血,乳腺腺泡壁固有结构受到破坏、乳腺腺泡壁增厚、炎性细胞浸润,乳腺组织损伤评分极显著升高(P<0.01),炎症因子(TNF-α、IL-1β和IL-6)浓度极显著升高(P<0.01),紧密连接蛋白(ZO-1、Occludin、Claudin-3)表达量极显著降低(P<0.01).低、中、高剂量组均显著地逆转了S.aureus诱导的上述指标的变化(P<0.05或P<0.01).与对照组相比,LYC组所有指标均无显著差异(P>0.05).LYC对S.aureus的MIC为128 μg/mL;亚抑菌浓度(16、32、64 μg/mL)LYC极显著抑制S.aureus溶血活性(P<0.01),并呈剂量依赖性地降低Hla蛋白表达(P<0.01);而8 μg/mL LYC对S.aureus溶血活性和Hla蛋白表达均无显著影响(P>0.05).结果表明,LYC通过调控宿主炎症反应和细菌毒力以缓解S.aureus诱导的乳腺炎,为天然产物靶向"宿主-病原体"互作而防控乳腺炎提供了新策略.

This study aimed to investigate the protective effects and underlying mechanisms of lycopene(LYC)on Staphylococcus aureus(S.aureus)-induced mastitis in mice.A total of 36 pregnant Kunming mice(6-8 weeks old)were randomly divided into six groups:Control,LYC,Model,Low-dose,Medium-dose,and High-dose groups.From mid-pregnancy(day 7 after vaginal plug detection),the LYC,Low-,Medium-,and High-dose groups were administered 50,12.5,25,and 50 mg/(kg·bw)of LYC by gavage once daily,respectively,while the Control and Model groups received an equal volume of corn oil.After 28 days of treatment,mastitis was induced by injecting S.aureus suspension(2×10⁸ CFU/mL)into the mammary ducts(Control group and LYC group received no treatment).After 24 hours,the mice were euthanized,and the anatomical changes in the mammary glands were observed.The fourth pair of mammary glands was collected for histopathological analysis by hematoxylin and eosin(H.E.)staining.Levels of inflammatory cytokines[tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),and interleukin-1β(IL-1β)]were measured using enzyme-linked immunosorbent assay(ELISA),and the expression of tight junction proteins[zonula occludens-1(ZO-1),Occludin,and Claudin-3]was evaluated by Western blot(WB).Additionally,the antibacterial activity of LYC against S.aureus was determined using the broth microdilution method to measure its minimum inhibitory concentration(MIC)and growth curve.Hemolytic assays and WB were used to assess S.aureus hemolytic activity and α-hemolysin(Hla)expression,respectively.The results showed that compared with the Control group,the Model group exhibited mammary tissue hemorrhage,destruction and thickening of alveolar wall structure,infiltration of inflammatory cells,a significantly higher tissue injury score(P<0.01),significantly elevated levels of TNF-α,IL-1β,and IL-6(P<0.01),and significantly decreased expression of ZO-1,Occludin,and Claudin-3(P<0.01).These pathological changes were significantly reversed by LYC treatment in all dose groups(P<0.05 or P<0.01).No significant differences were observed between the Control group and the LYC group across all indicators(P>0.05).The MIC of LYC against S.aureus was 128 μg/mL.Sub-inhibitory concentrations of LYC(16,32,and 64 μg/mL)significantly suppressed hemolytic activity(P<0.01)and dose-dependently reduced Hla expression(P<0.01),whereas 8 μg/mL LYC had no significant effect(P>0.05).In conclusion,LYC alleviates S.aureus-induced mastitis by modulating host inflammatory responses and bacterial virulence,providing a novel strategy for the prevention and control of mastitis through natural products targeting host-pathogen interactions.
