ELISA法与UPLC-MS/MS法检测水产品中喹诺酮类药物残留的比较分析OA
Comparative analysis of ELISA and UPLC-MS/MS methods for detecting quinolone residues in aquatic products
采用酶联免疫法(ELISA)和超高效液相色谱-串联质谱联用法(UPLC-MS/MS)检测市售6种水产品中14种喹诺酮类药物(QNs)残留的效果,旨在比较两种方法结果的差异性和相关性.其中,UPLC-MS/MS法参考国家标准GB/T 20366-2006进行检测分析,建立14种QNs药物标准曲线,外标法进行定量分析的结果与ELISA法比较.结果表明:ELISA法检测QNs类药物样品阳性检出率为78.57%,UPLC-MS/MS法检出率为64.29%,环丙沙星最高残留量在黄颡鱼头中为15.76 μg/kg,恩诺沙星UPLC-MS/MS法的阳性检出率为64.29%,且最高残留量高达2128.41 μg/kg.其中ELISA法样品检出率普遍高于UPLC-MS/MS法,说明ELISA法存在假阳性的情况.鉴于ELISA法分析速度快,样品前处理方法简单等优点,适合用于大批量的样品初筛,UPLC-MS/MS法准确度高,重现性好,可用于ELISA法阳性结果的验证和更精确的药物残留含量测定.本实验为水产品中监测QNs药物残留提供新的检测思路,可结合实际工作情况,选择更快更好的方法,为水产品中用药或水产养殖饲料添加剂的QNs类药物残留检测提供技术支持.
Enzyme-linked immunoassay(ELISA)and ultra-performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS)were used to detect the effects of 14 quinolones(QNs)in six commercially available aquatic products,aiming to compare the differences and correlations between the two methods.Among them,the detection conditions of the UPLC-MS/MS method were detected and analyzed with reference to the national standard GB/T 20366-2006,and the standard curves of 14 QNs drugs were established,and the quantitative analysis results of the external standard method were compared with the ELISA method.The results showed that the positive detection rate of QNs drug samples was 78.57%by ELISA,64.29%by UPLC-MS/MS,the highest residue of ciprofloxacin was 15.76 μg/kg in the head of yellow catfish,and the positive detection rate of enrofloxacin was 64.29%,and the highest residue was as high as 2128.41 μg/kg.In view of the advantages of ELISA method such as fast analysis speed and simple sample preparation method,it was suitable for the initial screening of large batches of samples,and the UPLC-MS/MS method had high accuracy and good reproducibility,which could be used for the verification of positive results of ELISA method and more accurate determination of drug residue content.This experiment provided a new detection idea for the monitoring of QNs drug residues in aquatic products,and could choose faster and better methods based on the actual working conditions,so as to provide technical support for the detection of QNs drug residues in aquatic products or aquaculture feed additives.
