目的 探讨健步汤抗骨质疏松症(OP)的作用.方法 18 只大鼠采取双侧卵巢摘除法构建OP模型,造模成功后随机分为模型组、仙灵骨葆组(0.27 g/kg)、健步汤组(15.75 g/kg),每组 6 只;另设空白组和假手术组,每组6 只.连续给药 90d后,采用Micro-CT检测骨微结构变化,ELISA法检测大鼠血清骨形成标志物PINP、骨吸收标志物CTX-1和炎症因子TNF-α、IL-6水平,流式细胞术检测大鼠股骨组织细胞凋亡率,RT-qPCR法检测大鼠股骨组织PI3K、Akt、PTEN mRNA表达.采取地塞米松诱导MC3T3-E1 细胞建立OP 体外模型,分为空白组、模型组和健步汤含药血清低、中、高浓度(健步汤-L、M、H)组(10%、15%、20%含药血清),干预结束后,采用CCK-8 法检测细胞增殖活性,流式细胞术检测细胞凋亡率,Western blot法检测细胞PI3K、p-PI3K、Akt、p-Akt、Bax、Bcl-2 蛋白表达,RT-qPCR法检测细胞PI3K、Akt、BGP、Runx2、COL1a1 mRNA表达.结果 健步汤能增加OP大鼠骨密度、骨体积分数和骨小梁数量(P<0.05),升高PINP水平(P<0.05),降低CTX-1、TNF-α、IL-6 水平(P<0.05),降低股骨组织中细胞凋亡率(P<0.05),降低PTEN mRNA表达(P<0.05),升高PI3K、Akt mRNA表达(P<0.05).健步汤含药血清能增加MC3T3-E1 细胞增殖活性(P<0.05),降低细胞凋亡率(P<0.05),升高p-PI3K、p-Akt、Bcl-2 蛋白表达(P<0.05),降低 Bax 蛋白表达(P<0.05),升高 PI3K、Akt、BGP、Runx2、COL1a1 mRNA 表达(P<0.05).结论 健步汤可能通过激活PI3K/Akt信号通路,促进MC3T3-E1 细胞增殖和活性,减少细胞凋亡,同时增加骨密度并减轻机体炎症反应,从而发挥抗骨质疏松症的作用.

AIM To investigate the anti-osteoporosis(OP)effects of Jianbu Decoction.METHODS Eighteen rats were selected to establish OP models by bilateral ovariectomy.After successful modeling,they were randomly divided into model group,Xianling Gubao group(0.27 g/kg)and Jianbu Decoction group(15.75 g/kg),with 6 rats in each group.The blank group and the sham operation group were also set up,with 6 rats in each group.After continuous administration for 90 days,Micro-CT was used to detect changes in bone microstructure.ELISA was employed to measure the serum levels of bone formation marker PINP,bone resorption marker CTX-1,and inflammatory factors TNF-α and IL-6 in rats.Flow cytometry was used to detect the apoptosis rate of femoral tissue cells.RT-qPCR was applied to determine the mRNA expressions of PI3K,Akt and PTEN in rat femoral tissue.Dexamethasone-induced MC3T3-E1 cells were used to establish the in vitro OP model,which was divided into blank group,model group,and low,medium and high concentration groups of Jianbu Decoction-containing serum(Jianbu Decoction-L,M,H groups)(10%,15%,20%drug-containing serum).After intervention,CCK-8 assay was used to detect cell proliferation activity.Flow cytometry was used to measure cell apoptosis rate.Western blot was performed to detect the protein expressions of PI3K,p-PI3K,Akt,p-Akt,Bax and Bcl-2.RT-qPCR was used to determine the mRNA expressions of PI3K,Akt,BGP,Runx2 and COL1a1in cells.RESULTS Jianbu Decoction could increase bone mineral density,bone volume fraction and trabecular number in OP rats(P<0.05),elevate PINP level(P<0.05),reduce the levels ofCTX-1,TNF-α and IL-6(P<0.05),decrease the apoptosis rate of cells in femoral tissue(P<0.05),downregulate PTEN mRNA expression(P<0.05),and upregulate PI3K and Akt mRNA expressions(P<0.05).Jianbu Decoction-containing serum could enhance the proliferation activity of MC3T3-E1 cells(P<0.05),reduce cell apoptosis rate(P<0.05),increase the protein expressions of p-PI3K,p-Akt and Bcl-2(P<0.05),decrease Bax protein expression(P<0.05),and upregulate the mRNA expressions of PI3K,Akt,BGP,Runx2 and COL1a1(P<0.05).CONCLUSION Jianbu Decoction may exert an anti-osteoporosis effect by activating the PI3K/Akt signaling pathway,promoting the proliferation and activity of MC3T3-E1 cells,reducing cell apoptosis,increasing bone mineral density and alleviating the body's inflammatory response.
