白竭散抑制JAK2/STAT3信号通路促进肛瘘大鼠创面愈合OA
Baijie Powder Promoted Wound Healing in Rats with Anal Fistula via Inhibiting the JAK2/STAT3 Signaling Pathway
目的 探讨白竭散对肛瘘大鼠术后创面愈合的影响.方法 采用细菌感染诱导法复制肛瘘大鼠模型.建模成功的 27 只大鼠随机分为模型组、白竭散组(1g/cm2)和贝复济组(150 IU/cm2),各 9 只,另取 10 只大鼠作为对照组.采用生理盐水清洁后喷洒相应药物,并以无菌纱布固定,每天 2 次,连续 2 周.观察大鼠创面愈合情况并计算愈合率,对创面水肿进行评分;采用苏木精-伊红(HE)染色,观察大鼠创面肉芽组织病理形态;采用酶联免疫吸附试验(ELISA)法检测大鼠创面肉芽组织白细胞介素 1β(IL-1β)、肿瘤坏死因子α(TNF-α)、白细胞介素 6(IL-6)、血管内皮生长因子(VEGF)、血管紧张素Ⅱ(AngⅡ)、血小板源性生长因子(PDGF)、胰岛素样生长因子1受体(IGF-1R)水平;采用免疫荧光法检测大鼠创面肉芽组织细胞凋亡情况并计算凋亡率;采用qRT-PCR法和Western blot法检测大鼠创面肉芽组织Janus激酶 2/信号传导及转录激活因子 3(JAK2/STAT3)相关mRNA和蛋白表达水平.结果 与模型组比较,白竭散组和贝复济组创面愈合率显著升高(P<0.05);创面水肿评分显著降低(P<0.05);IL-1β、TNF-α、IL-6 水平均显著降低(P<0.05);VEGF、AngⅡ、PDGF、IGF-1R水平均显著升高(P<0.05);创面肉芽组织细胞凋亡率显著降低(P<0.05);JAK2、STAT3 mRNA和蛋白表达水平均显著降低(P<0.05).结论 白竭散可减轻肛瘘大鼠机体的炎性反应,促进血管生成,抑制创面细胞凋亡,从而减轻创面水肿,促进术后创面愈合,其机制可能与抑制JAK2/STAT3 信号通路的激活有关.
Objective To investigate the effect of Baijie Powder on postoperative wound healing in rats with anal fistula.Methods The bacterial infection induction method was used to establish the anal fistula model rat.A total of 27 rats with successful modeling were randomly divided into the model group,the Baijie Powder group(1 g/cm2)and the Bovine Basic Fibroblast Growth Factor for External Use(Beifuji)group(150 IU/cm2),with 9 rats in each group,another 10 rats were selected as control group.After cleaning with normal saline,the corresponding drugs were sprayed on the wound,and fixed with sterile gauze,twice a day for two consecutive weeks.The wound healing in rats was observed,and the healing rate was calculated,and the wound edema was scored.Hematoxylin-eosin(HE)staining was used to observe the pathological morphology of granulation tissue in rat wound.Enzyme-linked immunosorbent assay(ELISA)method was used to detect the levels of interleukin-1 β(IL-1β),tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),vascular endothelial growth factor(VEGF),angiotensin Ⅱ(Ang Ⅱ),platelet-derived growth factor(PDGF)and insulin-like growth factor 1 receptor(IGF-1R)of granulation tissue in rat wound.Immunofluorescence method was used to detect the apoptosis cells of granulation tissue in rat wound and calculate the apoptosis rate.qRT-PCR and Western blot method were used to detect the mRNA and protein expression levels of Janus kinase 2/signal transducer and activator of transcription 3(JAK2/STAT3)of granulation tissue in rat wound.Results Compared with the model group,the wound healing rates in the Baijie Powder group and Beifuji group were significantly increased(P<0.05);the wound edema scores were significantly decreased,the levels of IL-1β,TNF-α and IL-6 were significantly decreased(P<0.05);the levels of VEGF,Ang Ⅱ,PDGF and IGF-1R were significantly increased(P<0.05);the apoptosis rates of granulation tissue in wound were significantly decreased(P<0.05);the expression levels of JAK2 and STAT3 mRNA and protein were significantly decreased(P<0.05).Conclusion Baijie Powder can alleviate the inflammatory reaction of rats with anal fistula,promote angiogenesis,inhibit wound cell apoptosis,reduce wound edema,and promote wound healing.Its mechanism may be related to the inhibition of activation of JAK2/STAT3 signaling pathway.
