为提高韶关翁源巴戟天总环烯醚萜得率,本试验采用纤维素酶辅助超声法提取,通过单因素试验与Box-Behnken响应面法设计试验优化工艺,并对总环烯醚萜的体外抗氧化活性进行了研究.结果表明:酶辅助超声提取巴戟天总环烯醚萜的最佳工艺条件为:酶量2%、酶解时间2 h、液料比33∶1(mL/g)、超声时间46 min、乙醇浓度39%.此参数下,总环烯醚萜得率为30.86%,与预测值的相对误差为0.16%.此外,巴戟天总环烯醚萜提取物具有一定的抗氧化活性,对超氧阴离子自由基清除的IC50为5.582 mg/mL,同时对钼酸铵有良好的还原能力.

To improve the total iridoids of Morinda officinalis from Shaoguan Wengyuan,the single test and Box-Behnken response surface method were designed by cellulase-assisted ultrasonic extraction,and the antioxidant activity in vitro was investigated.The results showed that the optimal extraction process conditions of total iridoids from Morinda officinalis with enzyme-assisted ultrasonic were:enzyme amount of 2%,enzyme digestion time of 2 h,liquid-to-feed ratio of 33∶1(mL/g),ultrasonic time of 46 min,and ethanol concentration of 39%.Under the condition,the yield of total iridoids was 30.86%,and the relative error to the predicted value was 0.16%.In addition,the total iridoids of Morinda officinalis showed certain antioxidant activity,with an IC50 of 5.582 mg/mL for scavenging superoxide anion radicals and a good reducing ability for ammonium molybdate.
