本研究旨在采用酶提和碱提两种方法从干燥罗汉果中提取多糖,并比较它们的多糖特征.通过单因素试验和正交试验,以罗汉果多糖的得率作为评价标准,研究酶提法中酶活力、温度、时间、pH以及碱提法中碱浓度、温度、时间、提取次数对罗汉果多糖提取的影响,优化制备条件.同时,还研究两种方法对α-葡萄糖苷酶和α-淀粉酶的抑制作用及抗氧化效果.结果显示:在酶提法中,最佳提取条件为酶活力3000 U、温度60 ℃、时间150 min、pH 5;在碱提法中,最佳提取条件为碱浓度1 mol/L、温度40 ℃、时间5 h、提取次数2次,碱提法的提取率最高,平均为5.99%,显著高于酶提法(2.18%).两种提取方法得到的多糖在总糖和糖醛酸含量上差异不大,酶提多糖的溶解度最低.碱提多糖在抑制α-葡萄糖苷酶和α-淀粉酶方面表现出更强的活性,这表明其在降血糖方面具有潜在的应用价值.不同方式提取的罗汉果多糖在清除DPPH自由基、羟基自由基和超氧阴离子方面表现出较好的能力,并具有明显的还原能力,表明罗汉果多糖具有较好的抗氧化活性.

The purpose of this study was to extracted polysaccharides from dried Siraitia grosvenorii using both enzymatic and alkaline extractions,and compared their polysaccharide profiles.Linvestigated the effects of enzyme activity,temperature,time,and pH in enzymatic extraction and alkaline concentration,temperature,time,and number of extractions in alkaline extraction,using the rate of polysaccharides of Siraitia grosvenorii as the evaluation criterion through the one-way test and orthogonal test,and optimized the preparation conditions.Meanwhile,their inhibitory effects on α-glucosidase,α-amylase and antioxidant effects were also investigated.The experimental results showed that in the enzyme extraction method,the optimal extraction conditions were enzyme activity of 3000 U,temperature of 60 ℃,time of 150 min,and pH of 5.In the alkaline extraction method,the optimal extraction conditions were alkali concentration of 1 mol/L,temperature of 40 ℃,time of 5 h,and the number of extraction times of 2.The alkaline extraction method yielded the highest extraction rate,which averaged 5.99%and was significantly higher than that of the enzyme extraction method(2.18%).The polysaccharides obtained by the two extraction methods did not differ much in total sugar and glucuronic acid content,and the solubility of the polysaccharides extracted by enzyme was the lowest.The alkali-extracted polysaccharides showed stronger activity in inhibiting α-glucosidase and α-amylase,which suggested their potential application in hypoglycemia.The Siraitia grosvenorii polysaccharides extracted in different ways showed better ability in scavenging DPPH radicals,hydroxyl radicals and superoxide anion,with obvious reducing ability,which indicated that Siraitia grosvenorii polysaccharides had better antioxidant activity.
