目的 采用生物信息学与动物实验结合法探究加味芎归汤(Jiawei Xionggui decoction,JWXG)改善高胆固醇血症相关性认知障碍(hypercholesterolemia-related cognitive impair-ment,HRCI)的治疗作用以及作用机制.方法 采用高分辨质谱检测JWXG入血成分;利用网络药理学、分子对接以及分子动力学模拟预测JWXG入血成分治疗HRCI的核心交集靶点,分析公开数据库中高胆固醇饮食(high cholesterol diet,HCD)模型小鼠脑组织转录组学差异基因;使用高胆固醇饲料构建HRCI小鼠模型,灌胃给药JWXG水煎液,试剂盒检测血清胆固醇含量,Morris水迷宫检测认知水平,HE染色评价神经病理损伤,Western blot检测关键蛋白表达,TUNEL染色评价凋亡神经元比例.结果 JWXG入血成分为黄芩素、黄芩苷、梓醇、白杨素、汉黄芩素和阿魏酸,入血成分核心交集靶点的KEGG和GO富集分析聚焦细胞凋亡途径,分子对接和分子动力学模拟显示黄芩苷、梓醇与Bax、Bcl-2结合力强,转录组学数据显示,Bax基因在HCD小鼠脑组织中高表达.动物实验结果表明,JWXG可明显降低HRCI小鼠血清胆固醇含量,改善学习记忆能力并减轻海马神经病理损伤,JWXG明显升高HRCI小鼠皮层Bcl-2/Bax蛋白比值、降低TUNEL阳性细胞数量.结论 JWXG通过拮抗神经细胞凋亡发挥神经细胞保护和改善HRCI的作用.

Aim To investigate the therapeutic effects of Jiawei Xionggui decoction(JWXG)on hypercholesterolemia-related cognitive impairment(HRCI)using an integrated approach combining bioin-formatics and animal experiments and the underlying mechanisms.Methods The blood-absorbed compo-nents of JWXG were identified by high-resolution mass spectrometry.Network pharmacology,molecular dock-ing,and molecular dynamics simulations were em-ployed to predict core targets of these components against HRCI.Transcriptomic data from brain tissues of high-cholesterol diet(HCD)model mice in public databases were analyzed to identify differentially ex-pressed genes.An HRCI mouse model was established using a high-cholesterol diet and treated with JWXG aqueous extract via gavage.Serum cholesterol levels were measured using assay kits,cognitive function was assessed with the Morris water maze,neural pathologi-cal damage was evaluated by HE staining,key protein expression was detected via Western blot,and neuro-nal apoptosis was examined by TUNEL staining.Re-sults The main blood-absorbed components of JWXG included baicalein,baicalin,catalpol,chrysin,wogo-nin,and ferulic acid.KEGG and GO enrichment analyses of core targets revealed a focus on the apopto-sis pathway.Molecular docking and dynamics simula-tions demonstrated strong binding affinities of baicalin and catalpol to Bax and Bcl-2.Transcriptomic data in-dicated high expression of the Bax gene in the brain tis-sues of HCD mice.Animal experiments showed that JWXG significantly reduced serum cholesterol levels,improved learning and memory abilities,and allevi-ated hippocampal neuropathological damage in HRCI mice.JWXG also markedly increased the Bcl-2/Bax protein ratio and decreased the number of TUNEL-positive cells in the cortex of HRCI mice.Conclu-sion JWXG exerts neuroprotective effects and ame-liorates HRCI by antagonizing neuronal apoptosis.
