鼠妇avu-miR-2866通过下调IGF1R逆转EMT抑制人肝癌细胞迁移侵袭OA
Armadillidium vulgare avu-miR-2866 reverses EMT to inhibit migration and invasion of human liver cancer cells by downregulating IGF1R
目的 探究鼠妇(Armadillidium vulgare Latreille)来源的微小RNA(microRNA,miRNA)avu-miR-2866对人肝癌细胞增殖、凋亡、迁移和侵袭的影响及分子机制.方法 Small RNA测序筛选鉴定出鼠妇特有miRNA;qRT-PCR检测avu-miR-2866在LX2、HepG2、MHCC97H、Hepa1-6细胞中表达水平;CCK-8、EdU染色、流式细胞术、细胞划痕实验和Transwell侵袭实验,检测肝癌细胞增殖、凋亡、迁移和侵袭的变化;Western blot检测上皮-间充质转化(epithelial-mesenchymal transition,EMT)相关蛋白E-cadherin、N-cadherin、Vimentin和MMP14的表达;生物信息学预测avu-miR-2866核心靶基因;Western blot检测靶基因蛋白的表达.结果 Small RNA测序筛选鉴定出avu-miR-2866;qRT-PCR显示avu-miR-2866在LX2、HepG2、MHCC97H、Hepa1-6细胞中的表达水平极低;功能实验显示,avu-miR-2866明显抑制HepG2、MHCC97H细胞增殖、迁移与侵袭,促进细胞凋亡;Western blot结果显示,avu-miR-2866 上调E-cadherin,下调N-cadherin、Vimentin和MMP14;生物信息学预测胰岛素样生长因子1受体(insulin-like growth factor 1 receptor,IGF1R)是avu-miR-2866的潜在靶基因,Western blot结果显示,avu-miR-2866 明显抑制IGF1R的表达.结论 鼠妇avu-miR-2866通过下调IGF1R逆转EMT进程,抑制人肝癌细胞迁移侵袭.
Aim To investigate the effects of the mi-croRNA(miRNA)avu-miR-2866,derived from Arma-dillidium vulgare Latreille(A.vulgare),on the prolif-eration,apoptosis,migration,and invasion of human liver cancer cells and to reveal the molecular mecha-nisms.Methods Small RNA sequencing approach was used to identify miRNAs specific to A.vulgare.The expression levels of avu-miR-2866 in various cell lines LX2,HepG2,MHCC97H and Hepa1-6 were measured by qRT-PCR.The effects of avu-miR-2866 on cell proliferation,apoptosis,migration,and inva-sion were evaluated using CCK-8 assay,EdU incorpo-ration assay,flow cytometry,wound healing assay,and Transwell invasion assay.The expressions of epithelial-mesenchymal transition(EMT)-related pro-teins E-cadherin,N-cadherin,Vimentin,and MMP14 were assessed using Western blot.The core target genes were predicted using bioinformatic analysis,and the target protein expression was further verified by Western blot.Results Avu-miR-2866 was identified through Small RNA sequencing.qRT-PCR analysis demonstrated that avu-miR-2866 expression was ex-tremely low in LX2,HepG2,MHCC97H and Hepa1-6 cells.Functional assays revealed that avu-miR-2866 significantly inhibited the proliferation,migration and invasion of HepG2 and MHCC97H cells,and pro-moted apoptosis.Western blot results showed that avu-miR-2866 upregulated E-cadherin and downregulated N-cadherin,Vimentin,and MMP14.Bioinformatics prediction suggested insulin-like growth factor 1 recep-tor(IGF1R)as a potential target gene of avu-miR-2866,and Western blot further confirmed that avu-miR-2866 significantly inhibited the expression of IGF1R.Conclusions Avu-miR-2866 derived from A.vulgare inhibits migration and invasion of human liver cancer cells by targeting IGF1R to reverse the EMT process,indicating its potential as a novel func-tional component of animal-derived traditional Chinese medicine in anti-metastatic therapy.
