丁香脂素对膝骨关节炎的治疗作用及机制研究OA

Investigating the therapeutic effects and mechanisms of syringaresinol on knee osteoarthritis via the TLR4/NF-κB signaling pathway

探讨丁香脂素(Syr)对膝骨关节炎(KOA)大鼠的软骨保护作用及TLR4/NF-κB通路调控机制.采用改良Hulth法构建SD大鼠KOA模型,30只大鼠随机分为假手术组、模型组、阳性药组(吲哚美辛,10 mg/kg)及Syr低、高剂量组(25、50 mg/kg),每组6只.结果显示,与模型组相比,Syr干预组大鼠运动功能显著改善(P<0.01),血清IL-1β、MMP13水平下降(P<0.01),软骨病理损伤减轻.蛋白免疫印迹及实时荧光定量聚合酶链式反应证实Syr可剂量依赖性下调软骨组织TLR4、P-NF-κB蛋白及mRNA表达(P<0.01),同时显著提升Ⅱ型胶原蛋白表达(P<0.01).机制研究表明,Syr通过抑制TLR4/NF-κB信号通路活化,有效缓解关节炎症并延缓软骨基质降解.上述结果为丁香脂素防治KOA和中医药现代化发展提供了理论依据.

This study investigated the chondroprotective effects of syringaresinol(Syr)on rats with knee osteoarthritis(KOA)and its regulatory mechanism via the TLR4/NF-κB pathway.A KOA model was established in SD rats using the modified Hulth method.Thirty rats were randomly divided into a sham-operated group,a model group,a positive drug group(indomethacin,10 mg/kg),and low-and high-dose Syr groups(25 and 50 mg/kg),with six rats in each group.The results showed that compared with the model group,Syr intervention significantly improved motor function(P<0.01),reduced serum levels of IL-1β and MMP13(P<0.01),and alleviated cartilage pathological damage.Western blot and real-time quantitative polymerase chain reaction confirmed that Syr dose-dependently downregulated the protein and mRNA expression of TLR4 and P-NF-κB in cartilage tissue(P<0.01),while significantly increasing the expression of type Ⅱ collagen(P<0.01).Mechanistic studies indicated that Syr effectively alleviated joint inflammation and delayed cartilage matrix degradation by inhibiting the activation of the TLR4/NF-κB signaling pathway.These findings provide a theoretical basis for the clinical application of syringaresinol in the prevention and treatment of KOA and for the modernization of Chinese medicine.
