扭肚藤多糖提取工艺优化及体外抗炎活性评价OA

Optimization of extraction process and anti-inflammatory activity in vitro evaluation of polysaccharides from Jasminum elongatum

以扭肚藤总多糖提取率为指标,以超声功率、超声时间(t)、提取温度(T)和液料比为考察因素,采用单因素和响应面实验优化扭肚藤多糖的提取工艺,并考察对脂多糖(LPS)诱导的RAW264.7细胞分泌炎症因子的影响.优化的扭肚藤多糖超声提取最佳工艺为t33 min,超声功率341 W,T62℃,液料比39 mL/g,提取率为(6.98±0.34)%.扭肚藤多糖以剂量依赖的方式,抑制LPS诱导的TNF-α、IL-1β和IL-6的生成,促进IL-10的产生.本文建立的扭肚藤多糖超声提取工艺稳定,方法可行;提取得到的扭肚藤多糖具有一定的体外抗炎活性,为扭肚藤的深入研究提供了依据.

To optimize the ultrasonic extraction process of polysaccharides from Jasminum elongatum(Bergius)Willd.and study its anti-inflammatory activity in vitro.The total polysaccharide yield from Jasminum elongatum served as the primary response indicator.Ultrasound power,time,extraction temperature,and liquid-solid ratio were selected as independent variables to optimize the extraction process using both single-factor experiments and response surface methodology(RSM).Additionally,the impact on inflammatory factors released by LPS-stimulated RAW264.7 cells was evaluated.The optimal ultrasonic extraction parameters for Jasminum elongatum polysaccharides was obtained as follows:ultrasonic time of 33 min,ultrasonic power of 341 W,extraction temperature of 62 ℃,liquid-solid ratio of 39 mL/g.Under these conditions,the polysaccharide yield reached(6.98±0.34)%.The polysaccharides from Jasminum elongatum exhibited a dose-dependent inhibited the LPS-induced production of TNF-a,IL-1β,and IL-6 while enhancing the generation of IL-10.The optimized ultrasonic extraction process of polysaccharides from Jasminum elongatum is stable and feasible.The polysaccharides from Jasminum elongatum demonstrated anti-inflammatory activity in vitro,which provides theoretical basis for the further research on Jasminum elongatum.
