养血补肾方调节CAV1/AQP1通路对形觉剥夺性近视大鼠TGF-β2表达、巩膜重塑及水转运的影响OA
Effects of Yangxue Bushen formula on transforming growth factor-β2 expres-sion,scleral remodeling and water transport in rats with form-deprivation myopia by regulating the caveolin-1/aquaporin 1 pathway
目的 探究养血补肾方调节小窝蛋白-1(CAV1)/水通道蛋白1(AQP1)通路对形觉剥夺性近视(FDM)大鼠转化生长因子-β2(TGF-β2)表达、巩膜重塑及水转运的影响.方法 收集SPF级雄性SD大鼠40只,取30只大鼠建立FDM模型,建模成功后将其分为FDM组、哌仑西平(PIR)组、养血补肾方组,每组各10只,另将10只未建模大鼠作为正常组.正常组与FDM组大鼠均不进行任何干预,PIR组每天早、晚给予PIR滴眼液干预,养血补肾方组大鼠灌胃养血补肾方干预.采用检影镜及诊断仪检测大鼠屈光度、眼轴长度,HE染色检测大鼠巩膜组织形态学变化,免疫组织化学染色检测大鼠巩膜组织中TGF-β2表达,Western blot检测大鼠巩膜组织中AQP1、CAV1蛋白相对表达水平,RT-PCR检测大鼠巩膜组织中天冬氨酸转运体(GLAST)、谷氨酸转运体1(GLT-1)mRNA相对表达水平.结果 与正常组比较,FDM组、养血补肾方组、PIR组大鼠屈光度、TGF-β2表达水平、AQP1蛋白、CAV1蛋白相对表达水平均降低,眼轴长度、GLAST mRNA、GLT-1 mRNA相对表达水平均升高(均为P<0.05);与FDM组比较,养血补肾方组、PIR组大鼠屈光度、TGF-β2表达水平、AQP1蛋白、CAV1蛋白相对表达水平均升高,眼轴长度、GLAST mRNA、GLT-1 mRNA相对表达水平均降低(均为P<0.05);与养血补肾方组比较,PIR组大鼠屈光度、TGF-β2表达水平、AQP1蛋白、CAV1蛋白相对表达水平均降低,眼轴长度、GLAST mRNA、GLT-1 mRNA相对表达水平均升高(均为P<0.05).HE染色结果显示,正常组大鼠视网膜各层结构无异常改变;FDM组大鼠视网膜变薄且神经节细胞层出现严重空泡样改变;养血补肾方组大鼠与PIR组大鼠视网膜厚度均增加,细胞排列基本规则,神经节细胞数量增多,各层细胞排列致密,其中以养血补肾方组大鼠表现最显著.结论 养血补肾方对FDM的治疗具有一定的积极效用,其机制可能与调节CAV1、AQP1、TGF-β2表达及平衡和改善巩膜重塑及水转运功能相关.
Objective To explore the effects of the Yangxue Bushen formula on transforming growth factor-β2(TGF-β2)expression,scleral remodeling,and water transport in rats with form-deprivation myopia(FDM)by regulating the caveolin-1(CAV1)/aquaporin 1(AQP1)pathway.Methods Forty SPF-grade male SD rats were collected.Thirty rats were selected to establish FDM models.After successful modeling,they were divided into the FDM group,the pirenzepine(PIR)group,and the Yangxue Bushen formula group,with 10 rats in each group.Additionally,10 rats that were not modeled were classified as the normal group.Rats in the normal group and the FDM group received no intervention.Rats in the PIR group were given PIR eye drops in the morning and evening every day for intervention,while rats in the Yangxue Bushen formula group were given the Yangxue Bushen formula by gavage for intervention.The refractive power and axial length of rats were detected by retinoscopy and diagnostic instruments.The morphological changes in the scleral tissue of rats were detected by hematoxylin-eosin(HE)staining.The TGF-β2 expression in the scleral tissue of rats was detected by immuno-histochemical staining.The relative expression levels of AQP1 and CAV1 in the scleral tissue of rats were measured by West-ern blot,and the relative mRNA expression levels of aspartate transporter(GLAST)and glutamate transporter 1(GLT-1)in the scleral tissue of rats were measured by real-time polymerase chain reaction.Results Compared with the normal group,the refractive power,the expression level of TGF-β2,and the relative expression levels of AQP1 and CAV1 proteins in the FDM group,the Yangxue Bushen formula group,and the PIR group decreased,while the axial length and the relative ex-pression levels of GLAST and GLT-1 mRNAs increased(all P<0.05).Compared with the FDM group,the refractive power,the expression level of TGF-β2,and the relative expression levels of AQP1 and CAV1 proteins in the Yangxue Bushen formu-la group and the PIR group increased,while the axial length and the relative expression levels of GLAST and GLT-1 mRNAs decreased(all P<0.05).Compared with the Yangxue Bushen formula group,the refractive power,the expression level of TGF-β2,and the relative expression levels of AQP1 and CAV1 proteins in the PIR group decreased,while the axial length and the relative expression levels of GLAST and GLT-1 mRNAs increased(all P<0.05).The results of HE staining showed that there were no abnormal changes in the structure of each layer of the retina in the normal group.In the FDM group,the reti-na of rats became thinner,and severe vacuolar changes occurred in the ganglion cell layer.The retinal thickness of rats in the Yangxue Bushen formula group and the PIR group increased,the cell arrangement was basically regular,the number of ganglion cells increased,and the cells in each layer were densely arranged.Among them,the rats in the Yangxue Bushen for-mula group showed the most significant performance.Conclusion The Yangxue Bushen formula has a positive effect on FDM,which may be related to the regulation of CAV1,AQP1,and TGF-β2 expression levels,and balance and improvement of scleral remodeling and water transport functions.
