电针风府、哑门穴治疗偏头痛模型大鼠的作用效果及机制研究OA
Effects and Mechanisms of Electroacupuncture at Fengfu and Yamen Acupoints in Migraine Model Rats
目的:探讨电针风府、哑门穴对偏头痛大鼠的疗效及作用机制.方法:实验分为正常组(A)、模型组(B)、电针组(C)、电针对照组(D).采用皮下注射硝酸甘油进行造模.各组给予对应的治疗7天后,开展行为学评分和分子实验.结果:行为学评分显示,相对模型组,经电针治疗的大鼠,挠头、爬笼等行为次数明显下降.免疫组织化学及免疫蛋白印迹实验结果均显示,给予电针治疗的偏头痛大鼠,在海马部位的5-HT1A表达升高(P<0.05),而TLR2、IL-1β表达下调(P<0.05);而与D组相比,C组在升高5-HT1A,下调TLR2、IL-1β的表达更有优势(P<0.05).结论:穴位有一定的特异性,给予穴位电针刺激,能减轻大鼠偏头痛的症状,其机制可能与上调5-HT1A表达,降低TLR2、IL-1β表达有关.
Objective:To investigate the efficacy and mechanism of electroacupuncture at Fengfu and Yamen acupoints in treating migraine rats.Methods:The experiment was divided into a normal group(A),model group(B),electroacu-puncture group(C),and electroacupuncture control group(D).The model was established by subcutaneous injection of nitroglycerin.After 7 days of corresponding treatment in each group,behavioral scoring and molecular experiments were conducted.Results:Behavioral scores showed a significant decrease in the number of head scratching and cage climbing behaviors in rats treated with electroacupuncture relative to the model group.The results of both immunohis-tochemistry and immunoprotein blotting experiments showed that migraine rats given electroacupuncture treatment had elevated 5-HT1A expression in the hippocampus(P<0.05),whereas TLR2 and IL-1β expression were downregu-lated(P<0.05).And compared with D,group C was more advantageous in elevating 5-HT1A and down-regulating the expression of TLR2 and IL-1β(P<0.05).Conclusion:Acupuncture points have certain specificity,and electroacupunc-ture stimulation given to acupuncture points can reduce the symptoms of migraine in rats,and the mechanism may be related to the up-regulation of the expression of 5-HT1A,and the down-regulation of the expression of TLR2 and IL-1β.
