参照国际人用药品注册技术协调会(ICH)的《Q3D(R2):元素杂质指导原则》(ICH Q3D)等相关指导原则,建立电感耦合等离子体-质谱(ICP-MS)法同时测定生脉注射液中 34 种重金属元素含量的测定方法.将供试品溶液微波消解后再稀释,经过ICP-MS法进行分析,通过加In、Sc为内标进行校正,等离子体输出功率为 1 150 W,采样深度为 8 mm,最大载气流速为 1.045 L/min,冷却气流速为 14 L/min,辅助气流速为 0.9 L/min,蠕动泵转速为40 r/min,进样系统为耐酸系统,取样锥及截取锥的型式为镍锥,手动采样,重复 3 次.结果发现:34 种重金属元素线性均良好(相关系数r均大于 0.99),平均回收率为 70%~120%,重复性的相对标准偏差(RSD)均小于 25%,中间精密度的RSD均小于25%.6 批生脉注射液中,34 种重金属元素含量结果均符合要求.上述方法便捷、快速、灵敏、准确,可进行生脉注射液中 34 种重金属元素浓度的检测,经过初步的方法学验证后,可更全面地评估生脉注射液的安全性,该方法可为注射剂更加严密的质量管理提供依据.

In accordance with the relevant guidelines of ICH Q3D(R2)Guideline for Elemental Impurities,an analytical method was established for the simultaneous determination of 34 heavy metal elements in Shengmai injection by ICP-MS.The test solution was ablated under microwave,then diluted and analyzed by ICP-MS method.The assay data were further calibrated by adding In and Sc as the internal standard.The plasma output power was set at 1 150 W,the sampling depth was 8 mm,the maximum carrier gas volume was 1.045 L/min,the cooling gas was 14 L/min and the auxiliary gas was 0.9 L/min.The peristaltic pump speed was 40 r/min,the feeding system was acid-resistant system,with nickel material for the sampling and interception cones.The sampling method was manual,and the assay was repeated 3 times.As a result,our method demonstrated excellent linearity for the 34 heavy metal elements(r>0.99),with the average recoveries ranging from 70%to 120%.In addition,both reproducibility RSD and the intermediate precision RSD were less than 25%.Our analysis of 6 batches of Shengmai injectionsshowed the 34 heavy metal elements met the requirements of ICH Q3D.Overall,we developed a convenient,rapid,sensitive and accurate method for the determination of the concentrations of 34 heavy metal elements in Shengmai Injections.Based on our preliminary methodological validation,the safety of Shengmai Injection can bebroadly monitored,and ourmethod can provide a basis for more rigorous quality management in this area.
