Large-scale production and efficacy of high-purity supramolecular elastin

本研究旨在开发超分子弹性蛋白的高效纯化技术,并系统评估其在细胞修复与皮肤护理中的生物学功效,为其在护肤领域的应用提供理论依据.通过镍柱亲和层析技术优化纯化工艺,蛋白自组装形成水凝胶提高蛋白稳定性,实现规模化生产.在细胞层面,通过增殖实验、划痕迁移实验和细胞黏附实验(0.5~10 g/L质量浓度梯度)评估超分子弹性蛋白对人皮肤成纤维细胞的作用;在人体皮肤测试中,通过皮肤光泽度分析、水分含量检测和弹性测定验证其护肤功效.在蛋白纯化得率保持在 97%的基础上,成功制备纯度高达 98.5%的超分子弹性蛋白,以其形成的水凝胶在常温储存 12 个月才出现降解.细胞实验结果显示:0.5 和1.0 μg/mL的超分子弹性蛋白分别促进细胞增殖 23.91%和 57.49%,而 10g/L水凝胶使细胞黏附率提升 82.22%.人体测试结果表明:使用 28d后,皮肤光泽度提升 29.2%,水分含量增加 27.5%,皮肤弹性改善 11.9%(P<0.01).建立的规模化纯化工艺可稳定制备高纯度超分子弹性蛋白,其通过促进成纤维细胞活性显著改善皮肤生理指标.实验数据证实该蛋白在保湿、抗衰等护肤领域具有重要应用价值,为开发新型生物活性护肤品提供了科学依据.

This study aims to develop efficient purification techniques for supramolecular elastin and systematically evaluate its biological efficacy in cell repair and skin care,providing a theoretical basis for its application in the skincare field.The purification process was optimized through nickel column affinity chromatography for large-scale production,and the protein self-assembled into hydrogels to enhance stability.With regards to the effects on human skin fibroblasts,various cellular methods were utilized for the assessment,including proliferation assay,scratch migration assay,and cell adhesion assay(concentration gradient of 0.5-10 g/L).In addition,the human skincare efficacy was verified through skin gloss analysis,moisture content detection,and elasticity measurement.Starting from the protein purified under optimized conditions,supramolecular elastin was successfully prepared,giving excellent purity(up to 98.5%).More importantly,the formed hydrogel demonstrated great stability at room temperature,without any visible degradation within 12 months of storage.At cellular level,supramolecular elastin could greatly promote cell proliferation,with 23.91%and 57.94%improvement at concentrations of 0.5 μg/mL and 1 μg/mL,respectively.Cell adhesion could also be improved,with 82.22%enhancement by treatment with 10 g/L hydrogel.Human tests were next conducted for the use of 30 days,and positive results were obtained at the end of the studies.In details,skin gloss increased by 29.2%,moisture content rose by 27.5%,and skin elasticity improved by 11.9%(P<0.01).In summary,a large-scale purification process was established for reliable production of high-purity supramolecular elastin,which demonstrated significant improvement of skin physiological indicators by promoting fibroblast activity.Experimental data also showed the potential application of this protein in skincare areas such as moisturizing and anti-aging,providing scientific ground for the development of new bioactive skincare products.
