目的 探讨葡萄籽原花青素是否对孕期应激的SD大鼠子代海马星形胶质细胞及认知功能有影响.方法 SD孕鼠被随机分为对照组、应激组和葡萄籽原花青素组,每组6只.应激组在孕15~20 d给予每天3次限制性应激,放置于透明塑料容器中限制活动45 min/次,葡萄籽原花青素组在孕15~20 d给予限制性应激,自应激开始至子代出生20 d的母代和21~60日龄的子代均饲喂葡萄籽原花青素(200 mg/kg),对照组正常饲养.60日龄的子代通过旷场、高架十字迷宫、Y迷宫评估认知功能;免疫组化和免疫荧光染色观察子代海马区星形胶质细胞形态、密度等改变;qRT-PCR检测子代海马中IL-1β、IL-6和TNF-α等促炎因子mRNA的表达水平,Western blot检测子代海马中星形胶质细胞标志物(GFAP)和NF-κB表达水平.结果 行为学结果表明,与对照组比较,应激组子代在旷场运动总路程和中央区路程明显减少(P<0.01),在高架十字迷宫开放臂停留时间百分比和进入开放臂次数百分比明显降低(P<0.001),在Y迷宫新异臂运动时间和运动距离百分比明显降低(P<0.01),表现为认知功能障碍;在葡萄籽原花青素组中,应激组的上述异常行为得到改善(P<0.05).应激组子代海马IL-1β、IL-6和TNF-α的mRNA水平较对照组升高(P<0.001),葡萄籽原花青素组相较于应激组上述指标的mRNA水平下降(P<0.05).免疫组化和免疫荧光结果显示,相比于对照组,应激组子代海马区星形胶质细胞密度降低、胞体变小、突起变短;与应激组相比,葡萄籽原花青素组子代海马区星形胶质细胞密度增加、形态恢复正常.Western blot检测结果显示,相比于对照组,应激组子代海马区GFAP表达降低(P<0.01),NF-κB表达升高(P<0.001);与应激组相比,葡萄籽原花青素组GFAP蛋白表达升高(P<0.05),NF-κB蛋白表达降低(P<0.01).结论 葡萄籽原花青素通过抑制NF-κB信号通路减轻孕期应激所致大鼠子代海马区星形胶质细胞损伤,保护子代认知功能.

Objective To investigate the effects of grape seed proanthocyanidins on hippocampal astrocyte and cognitive performance in the offsprings of SD rats exposed to prenatal stress.Methods Pregnant SD rats were randomized into control group,stress group,and grape seed proanthocyanidin(GSP)group.At gestational days 15-20,the prenatal rats subjected to restraint stress three times per day in a transparent plastic container that restricted movement for 45 min per session in stress group and GSP group.Based on this,the dam from GD15 to postnatal day 20(PD20)and the offsprings from PD21-60 in GSP group were given orally GSP 200 mg/(kg·d).The rats in control group were fed normally.At PD60,the open-field,elevated plus-maze,and Y-maze tests were used to evaluate cognitive performance in offsprings.Hippocampal astrocytic morphology and density were assessed by immunohistochemistry and immunofluores-cence.The mRNA levels of pro-inflammatory cytokines IL-1β,IL-6,and TNF-α were detected in hippocampus of offsprings by qRT-PCR,while the protein expressions of astrocyte marker(GFAP)and NF-κB were determined by Western blot.Results Compared with control group,total locomotor distance and central zone distance in the open-field test significantly decreased in offsprings in stress group(P<0.01),the percentages of time spent in and entries into the open arms also decreased in the elevated plus-maze test(P<0.001),and the percentages of total locomotion time and distance spent in the novel arm in the Y-maze test decreased(P<0.01).In GSP group,the behavioral deficits induced by stress were significantly ameliorated(P<0.05).The mRNA levels of IL-1β,IL-6,and TNF-α in hippocampus in stress group were significantly higher than in control group(P<0.001);these levels were significantly lower in GSP group than in stress group(P<0.05).Immunohistochemical and immunofluorescent results showed that PS offspring exhibited the reduction of astrocytic density,smaller somata,and shorter processes in the hippocampus compared with controls;GSP restored astrocytic density and normalized morphology.Compared with control group,hippocampal GFAP level significantly reduced in offsprings(P<0.01),and NF-κB expression significantly increased(P<0.001);compared with stress group,hipopocampal GFAP level significantly increased in GSP group(P<0.05),and NF-κB protein expression decreased(P<0.01).Conclusion Grape seed proanthocyanidins may alleviate PS-induced hippocampal astrocytic damage and protect cognitive function of the offsprings by inhibit-ing the NF-κB signaling pathway.
