靶向脂肪酸代谢重塑肿瘤免疫微环境:机制与免疫治疗新策略OA
Targeting Fatty Acid Metabolism to Reshape the Tumor Immune Microen-vironment:Mechanisms and Novel Immunotherapy Strategies
肿瘤的发生和进展与其微环境中多种因素的相互作用密不可分,其中脂肪酸代谢扮演着重要的角色.现有证据表明,脂肪酸不仅为肿瘤细胞的增殖和转移提供生物能量和生物合成前体,还可调控免疫细胞的功能状态与表型极化,重塑抑制性的肿瘤免疫微环境.尤其是脂肪酸的代谢途径及其衍生物在肿瘤的免疫逃逸、免疫耐药方面的作用,是目前的研究重点和热点.然而对脂肪酸代谢与肿瘤免疫微环境之间的关联仍缺乏充分的理解,特别是靶向代谢和免疫的联合治疗策略的探索.本文旨在分析脂肪酸代谢如何通过调节免疫细胞的功能状态以及改变肿瘤免疫微环境,从而影响肿瘤的免疫治疗效应.并总结当前的研究进展以及目前潜在的治疗策略,以期为肿瘤免疫治疗新策略的开发提供理论基础.
The development and progression of tumors are closely associated with the interactions of multiple factors with-in their microenvironment,where fatty acid metabolism plays a pivotal role.Accumulating evidence indicates that fatty acids not only supply bioenergy and biosynthetic precursors for tumor cell proliferation and metastasis,but also regulate the func-tional states and phenotypic polarization of immune cells,thereby reshaping the immunosuppressive tumor immune microenvi-ronment(TIME).Particularly,fatty acid metabolism and its derivatives are now central to research on tumor immune eva-sion and resistance mechanisms.However,the relationship between fatty acid metabolism and TIME remains poorly under-stood,especially regarding the exploration of combined therapies targeting both metabolism and immunity.This study aims to analyze how fatty acid metabolism influences tumor immune responses by modulating immune cell function and modifying TIME.It summarizes current research advancements and potential therapeutic strategies,providing theoretical foundations for developing novel immunotherapy approaches.
