基于生物信息学分析苍术抑菌作用研究OA

Antibacterial Effects of Cangzhu(Atractylodis Rhizoma)Based on Bioinformatics Analysis Experiments

目的 明确苍术抑菌主要部位及对各菌的抑菌作用.方法 运用数据库对苍术成分、靶点及抑菌相关靶点进行预测及筛选;利用DAVID数据库进行基因本体(GO)功能富集分析和京都基因与基因组百科全书(KEGG)通路富集分析;使用Cytoscape 3.10.2软件绘制"疾病-通路-靶点-成分-药物"网络图;采用Autodock软件进行分子对接;选择金黄色葡萄球菌、粪肠球菌、铜绿假单胞菌、鲍曼不动杆菌、表皮葡萄球菌、痤疮丙酸杆菌、糠秕马拉色菌进行体外抑菌实验验证.结果 共筛选得到苍术有效成分34个,成分靶点355个,抑菌靶点3678个,潜在治疗靶点200个;潜在抑菌成分与核心靶点均具有良好的结合能力,其中排名靠前的成分均为挥发油中有效成分.体外实验验证发现苍术挥发油对金黄色葡萄球菌、粪肠球菌、痤疮丙酸杆菌、糠秕马拉色菌抑制作用较好,表现为中度敏感;对表皮葡萄球菌抑制作用较弱,表现为低度敏感;对铜绿假单胞菌与鲍曼不动杆菌无抑制作用.结论 苍术主要抑菌部位为挥发油,其中对糠秕马拉色菌的抑制作用最强,对金黄色葡萄球菌和痤疮丙酸杆菌抑制作用较强,对表皮葡萄球菌抑制作用较弱,对铜绿假单胞菌与鲍曼不动杆菌无抑制作用.

Objective To clarify the main antibacterial parts of Cangzhu(Atractylodis Rhizoma)and its antibacterial effects on various bacteria.Methods The components,targets and antibacterial targets of Cangzhu(Atractylodis Rhizoma)were predicted and screened by database.DAVID database was used for gene ontology(GO)function enrichment analysis and Kyoto Gene and Genome Database(KEGG)pathway enrichment analysis.Cytoscape 3.10.2 software is used to map"disease-pathway-target-ingredient-drug".Using Autodock software for molecular docking;Staphylococcus aureus,Enterococcus faecalis,Pseudomonas aeruginosa,Acinetobacter baumannii,Staphylococcus epidermidis,Propionibacterium acnes and Malazzae furfur were tested in vitro to evaluate the inhibitory effect.Results A total of 34 active components,355 targets,3678 antibacterial targets and 200 potential therapeutic targets were obtained through network pharmacological screening.Potential antibacterial components and core targets have good binding ability,among which the top-ranked components are all effective components of volatile oil,so it is speculated that volatile oil is the main part of Cangzhu(Atractylodis Rhizoma)to exert antibacterial effect,and verified by in vitro experiments.Staphylococcus aureus,Enterococcus faecalis,Propionibacterium acens and Malassezia furfur were moderately sensitive to the volatile oil of Cangzhu(Atractylodis Rhizoma),and were low sensitive to Staphylococcus epidermidis.It had no inhibitory effect on Pseudomonas aeruginosa and Acinetobacter baumannii.Conclusion The results of network pharmacology and molecular docking technology showed that the main antibacterial part of Cangzhu(Atractylodis Rhizoma)was volatile oil.In vitro antibacterial experiment showed that the essential oil of Cangzhu(Atractylodis Rhizoma)had good antibacterial effect,among which the inhibitory effect on Malassezia furfur was the strongest,Staphylococcus aureus and Propionibacterium acnes were stronger.The inhibitory effect on Staphylococcus epidermidis was weak.It had no inhibitory effect on Pseudomonas aeruginosa and Acinetobacter baumannii.
