目的 改良现有胶质母细胞瘤类器官(GBO)的构建方法,探索其评估新型治疗方法效果的可行性.方法 收集术后病理分级WHOⅣ级的胶质母细胞瘤(GBM)标本,构建不同患者来源的肿瘤类器官和原代细胞,比较其与亲本肿瘤的相似性,并开展GBO用于评估电场治疗效果的体外实验.结果 18 例GBM标本中,共15 例构建成功,同时保留了亲本肿瘤的结构特征,能长期培养并用于实验.构建过程中能同时获得GBM原代细胞,在免疫荧光染色中表达CD133.体外实验证实电场治疗能够抑制GBO生长.结论 本研究改良一种能低成本快速构建GBO的方法,用于评估电场治疗的效果.

Objective To improve the method of establishing glioblastoma organoids(GBO)and explore the feasibility of evaluating the effect of new treatments.Methods The postoperative pathological tissues of glioblastoma(GBM)diagnosed as WHO grade Ⅳ were collected,and the tumor organoids and primary cells from different patients were established and compared with the parental tumors,and the GBO were used to evaluate the effect of tumor-treating field treatment in vitro.Results Fifteen of the eighteen GBM tissues were successfully established and retained the structural characteristics of the parental tumors,which could be cultured for a long time and used for experiments.Primary GBM cells were obtained during the process,and expressed CD133 in immunofluorescence staining.In vitro experiments confirmed that TTF treatment could inhibit the growth of GBO.Conclusions This study improved a low-cost method to rapidly establish GBO to evaluate the effect of tumor-treating field treatment.
