基于慢病毒包装系统,运用多质粒共转染技术,将相关基因导入含有特异性核酸序列的宿主细胞中进行包装,同时在外壳表达对应的刺突蛋白,制备了人类免疫缺陷病毒(HIV-1)假病毒标准物质.采用数字聚合酶链式 反 应(PCR)方 法 定 值,假 病 毒 标 准 物 质 GAG 基 因 拷 贝 数 浓 度 的 定 值 及 扩 展 不 确 定 度 为(300.12±0.54)×104 copies/mL(k=2).利用假病毒侵染依赖刺突蛋白与宿主细胞受体的识别,添加中和抗体干扰二者结合可抑制假病毒侵染这一特性,通过观察不同浓度中和抗体对荧光标记表达的抑制程度,以Reed-Muench两式法计算ZPD50,实现中和抗体效价的评估.

Based on lentiviral packaging system,the co-transfection technology of multi-plasmid was used to deliver the relevant genes into host cells containing specific nucleic acid sequences,and the human immunodeficiency virus(HIV-1)pseudovirus reference materials were fabricated.The corresponding spike protein was expressed on the outer shell.The standard value of copy number and expanded uncertainly of the reference materials was(300.12±0.54)×104 copies/mL(k=2)by digital polymerase chain reaction(PCR).Taking advantage of the characteristic that the pseudovirus infects by the recognition between the spike protein and the receptor of host cell,the binding could be interfered by adding neutralizing antibodies to inhibit the infection of the pseudovirus.By observing the inhibition degree of neutralizing antibodies at different concentrations on the expression of fluorescence,the Reed-Muench two-formula method was used to calculate the ZPD50 and evaluated the potency of neutralizing antibodies.
