山西省玉米大斑病菌生理小种、交配型及遗传多样性分析OA
Analysis of Physiological Race,Mating Type and Genetic Diversity of Exserohilum turcicum in Shanxi Province
为明确山西省玉米大斑病菌(Exserohilum turcicum)生理小种、交配型组成情况及病原菌遗传多样性,本研究采用Ht单基因鉴别寄主鉴定供试菌株的生理小种,通过交配型特异性引物测定供试菌株的交配型,并利用简单重复序列(SSR)分子标记技术对供试菌株进行遗传多样性分析.结果表明,山西省玉米大斑病菌共有 16 个(123N、123、23N、13N、12N、3N、2N、1N、23、13、12、N、3、2、1、0)生理小种,其中123N号和13N号生理小种为山西省优势小种.山西省玉米大斑病菌有Aa交配型、A交配型和a交配型3种,其中a交配型为主要交配类型.利用SSR标记分析了 52株玉米大斑病菌的遗传多样性,结果显示,6对SSR引物可扩增出40条多态性条带,多态性条带比例为100%.当遗传相似系数为0.69时,可将供试菌株划分为10个类群.分子方差分析结果表明,SSR类群的划分与地理来源和生理小种无明显相关性,与交配型之间存在较为明显相关性.本研究结果为山西省玉米大斑病菌的遗传多样性提供了信息,可为该病害的科学防治提供理论依据.
To clarify the physiological races,mating type composition,and genetic diversity of Exserohilum turcicum in Shanxi Province,the Ht single gene differential hosts were employed to determine the physiological races of the tested isolates,the mating type specific primers were utilized to determine their mating types,and genetic diversity was analyzed using SSR molecular markers in this study.The results showed that 16 physiological races of E.turcicum(123N,123,23N,13N,12N,3N,2N,1N,23,13,12,N,3,2,1,0)were identified in Shanxi Province,with 123N and 13N being the dominant races.Three mating types of Aa,A,and a were detected,with a being the most prevalent.SSR analysis of 52 E.turcicum isolates using 6 SSR primers yielded 40 polymorphic bands,achieving a polymorphism rate of 100%.Based on a genetic similarity coefficient of 0.69,the tested isolates were clustered into 10 distinct groups.Molecular variance analysis revealed that SSR-based classification exhibited no strong correlation with geographical origin or physiological race but showed a significant association with mating types.These findings provide valuable insights into the genetic diversity of E.turcicum in Shanxi Province and offer a theoretical foundation for effective management of northern corn leaf blight infected by Exserohilum turcicum.
