富含脯氨酸伸展蛋白样受体激酶(PERK)在植物生长、发育及胁迫响应中发挥关键作用.为了阐明PERK基因家族在药用植物丹参(Salvia miltiorrhiza)中的功能,本研究对其进行了全面的全基因组鉴定与分析,包括具体分析该家族成员的理化性质、染色体定位、共线性关系、系统发育关系、保守基序及顺式作用元件,并利用转录组测序和实时荧光定量PCR(qRT-PCR)技术,检测SmPERK基因在不同组织及水杨酸(SA)、脱落酸(ABA)处理和黄瓜花叶病毒(CMV)侵染下的表达模式.结果表明,丹参基因组中共鉴定出20个SmPERK基因,它们不均等地分布于6条染色体上.共线性分析表明,基因复制事件是该家族成员扩张的主要驱动力.表达谱分析显示出明显的组织表达特异性,如SmPERK12在花中高表达,而SmPERK11和SmPERK14则在根和茎中优势表达.多个SmPERK基因的表达对SA和ABA处理有响应,其中SmPERK14对ABA处理表现出极强的诱导反应.被CMV侵染后,多数家族成员的表达发生改变,特别是SmPERK13和SmPERK14表达显著上调.本研究系统地揭示了丹参SmPERK基因家族的结构特征与表达规律,阐明了其成员在组织分化、激素信号转导及抗病毒防御中可能发挥着不同作用.研究结果筛选出SmPERK13和SmPERK14等关键候选基因,为深入理解丹参PERK基因家族介导的生长发育与环境适应性调控网络提供了重要的理论依据和基因资源.

Proline-rich extensin-like receptor kinase(PERKs)play critical roles in plant growth,development,and stress responses.To elucidate the functions of the PERK gene family in medicinal plant Salvia miltiorrhiza,we performed a comprehensive genome-wide identification and analysis,such as chromosomal distribution,collinear relationships,phylogenetic relationships,conserved motifs,and cis-acting elements.Furthermore,expression patterns of SmPERK genes across different tissues and under salicylic acid(SA),abscisic acid(ABA),and cucumber mosaic virus(CMV)stress were investigated using transcriptome sequencing and quantitative real-time PCR(qRT-PCR).We identified 20 PERK genes,which were unevenly distributed across six chromosomes.Synteny analysis suggested that gene duplication events were the primary drivers of SmPERK family expansion.Expression profiling revealed distinct tissue-specific patterns:SmPERK12 was highly expressed in flowers,whereas SmPERK11 and SmPERK14 showed preferential expression in roots and stems.Several SmPERK genes responded to SA and ABA treatments,with SmPERK14 exhibiting particularly strong induction by ABA.Upon CMV infection,the expression of most family members was altered,with SmPERK13 and SmPERK14 being significantly upregulated.This study systematically characterizes the SmPERK gene family,clarifying its potential roles in tissue differentiation,hormone signaling,and antiviral defense.The identification of key candidate genes,particularly SmPERK13 and SmPERK14,provides a significant theoretical basis and valuable genetic resources for further understanding the regulatory network governing growth,development,and environmental adaptation mediated by the PERK gene family in S.miltiorrhiza.
