苯丙氨酸解氨酶(PAL)是苯丙烷代谢的关键限速酶,在植物生长发育和次生代谢等方面具有重要作用.为明确PALs家族在魔芋生长发育中的作用,本研究利用生物信息学方法对AkPALs家族成员的理化特性、系统发育树、基序及保守结构域、顺式元件等进行分析,同时比较了 AkPALs在魔芋不同器官、球茎的不同生长阶段及镧诱导下的表达模式.结果显示,7个魔芋AkPALs家族基因分布在5条染色体上,编码212~759个氨基酸,分布在3个分枝中,与单子叶植物亲缘关系较近.AkPALs家族启动子区域富集光响应、激素响应及逆境响应元件.结合转录组数据与镧诱导后的实时荧光定量PCR(qRT-PCR)分析,筛选出与魔芋球茎生长发育及其生物碱合成代谢密切相关的AkPAL4/5.本研究结果为魔芋AkPALs基因及其生物学功能的深入研究提供了理论参考.

Phenylalanine ammonia lyase(PAL)is indispensable in plants as the primary rate-limiting enzyme of phenylpropanoid metabolism,which underpins both growth and secondary metabolic pathways.To investigate the functions of PAL gene in the growth and development of Amorphophallus konjac,we conducted comprehensive analyses of AkPAL family members,including physicochemical properties,phylogenetic tree,conserved motifs/domains,and promoter characteristics.Additionally,expression levels of AkPALs across various tissues and during corm growth stages of A.konjac were investigated.Quantitative real-time PCR(qRT-PCR)was utilized to assess the expression of AkPALs following lanthanum induction.The results identified 7 AkPAL genes distributed across 5 chromosomes,encoding proteins of 212-759 amino acids that clustered into three phylogenetic clades with closer relationships to monocots.The protein secondary structure composition was predominantly characterized by α-helical segments and random coil conformations.Promoter regions exhibited significant enrichment of light,hormone,and stress-responsive cis-elements.Moreover,integrated transcriptomic and qRT-PCR analyses showed that AkPAL4/5 play significant roles in regulating both corm development and alkaloid biosynthesis in A.konjac.These results provide a theoretical reference for further studies on AkPALs genes and their biological functions in A.konjac.
