投喂配合饲料和冰鲜鱼对双须骨舌鱼生长性能、肠道组织结构和微生物及血清生化指标的影响OA
Effects of artificial compound feed and frozen fresh fish on growth performance,intestinal morphology,microbiota and serum biochemical indexes of Osteoglossum bicirrhosum
为比较摄食配合饲料和冰鲜鱼的双须骨舌鱼(Osteoglossum bicirrhosum)的生长性能、肠道组织结构、肠道微生物及血清生化指标的差异,分别以配合饲料(ACF组)和冰鲜鱼(FFF组)投喂双须骨舌鱼 70 d,测定其生长性能、肠道组织结构、肠道微生物菌群及血清生化指标.结果显示:1)ACF组的末体重、特定生长率、增重率和饲料系数均显著低于FFF组(P<0.05).2)ACF组双须骨舌鱼肠道组织结构存在损伤情况.3)通过对双须骨舌鱼肠道微生物菌群16S rRNA基因进行高通量测序和生物信息学分析,在0.97相似度下对样本细菌序列数据进行聚类,ACF组和FFF组6个样品数据共产生1 262个OTUs,其中FFF组和ACF组各有652和533个特有OTUs,共有OTUs77个;FFF组的Chao1指数、香农指数(Shannon)、辛普森指数(Simpson)以及OTU数量均高于ACF组,表明FFF组双须骨舌鱼肠道微生物多样性更加丰富.4)两组双须骨舌鱼肠道微生物菌群在门水平共有优势菌为变形菌门(Proteobacteria)和厚壁菌门(Firmicutes);相比FFF组,ACF组中变形菌门的相对丰度降低,厚壁菌门的相对丰度增加,多出了梭杆菌门(Fusobacteria)(43%);属水平共有的优势菌为爱德华氏菌属(Edwardsiella)、邻单胞菌属(Plesiomonas)、罗姆布茨菌(Romboutsia)和梭状芽孢杆菌属(Clostridium_sensu_stricto_1);相较FFF组,ACF组中爱德华氏菌属和邻单胞菌属的相对丰度降低,梭状芽孢杆菌属相对丰度升高,且增加了特异的鲸杆菌属(Cetobacterium)(43%).5)相较FFF组,ACF组双须骨舌鱼血清脂肪酶活性显著降低(P<0.05),但氧化氢酶、超氧化物歧化酶活性及甘油三酯、总蛋白含量差异不显著(P>0.05).结果表明:冰鲜鱼更适合双须骨舌鱼生长需求,配合饲料破坏了双须骨舌鱼肠道健康.研究结果可为丰富双须骨舌鱼饲料营养以及健康养殖提供基础资料.
Osteoglossum bicirrhosum is native to the Amazon River basin in Brazil and has large silver scales and a flat side.Due to its beautiful swimming posture and dignified posture,it is loved by many aquarium enthusiasts.At present,when raising O.bicirrhosum,it is usually fed with frozen fresh fish,causing resource waste and water pollution,and long-term consumption of frozen fresh fish may cause diseases in O.bicirrhosum.Artificial compound feed has the advantages of comprehensive nutrition,high safety,low feeding cost,and low pollution to water environment.This study investigated the effects of artificial compound feed(ACF)and frozen fresh fish(FFF)on the growth performance,intestinal microbiota and serum biochemical indexes of O.bicirrhosum,the culture experiment was carried out for 70 days.The results showed as follows:1)The final body weight,specific growth rate,weight gain rate and feed conversion ratio of the ACF group were significantly lower than those of the FFF group(P<0.05).2)There was damage to the intestinal morphology of O.bicirrhosum in the ACF group.3)High-throughput sequencing of 16S rRNA genes and bioinformatics analyses in the intestinal microbiotain of O.bicirrhosum and cluster of the sample bacterial sequence data with a similarity of 0.97 showed that there were 1 262 OTUs in the two groups,652 and 533 unique OTUs in the FFF group and the ACF group respectively,with a total of 77 OTUs.4)Proteobacteria and Firmicutes are the common dominant bacteria at the phylum level.When compared to the FFF group,the ACF group reduced the relative abundance of Proteobacteria,increased the relative abundance of Firmicutes,and increased the specific fusobacteria(43%).The dominant bacteria at the genus level were Edwardsiella,Plesiomonas,Romboutsia and Clostridium_sensu_stricto_1.Compared with FFF group,the ACF group reduced the relative abundance of Edwardsiella,Plesiomonas,increased the relative abundance of Clostridium_sensu_stricto_1,and increased the specific Cetobacterium(43%).5)Compared with the FFF group,the ACF group significantly reduced the activity of serum lipase(P<0.05),but had no significant effect on the activities of catalase,superoxide dismutase,triglyceride and total protein(P>0.05).The results of this study showed that FFF was more suitable for the growth needs of O.bicirrhosum,and ACF destroyed the intestinal health of O.bicirrhosum.The research results can provide basic information for enriching the nutritional value of ACF for O.bicirrhosum and promoting healthy aquaculture.
