两株鸡源鸭疫里默氏菌的分离鉴定及OmpA基因序列分析OA
Isolation,Identification and OmpA Gene Sequence Analysis of Two Strains of Riemerella anatipestifer from Chicken
为了鉴定从肉种鸡输卵管干酪样物和其孵化种蛋的卵黄中分离到的2株相似病原菌,对这2株分离菌进行革兰氏染色、血清型鉴定、药敏试验和OmpA基因分析,并将分离菌进行SPF鸡致病性试验.结果表明:从输卵管和卵黄中分离的菌株均为1型鸭疫里默氏菌(Riemerella anatipestifer,RA);2株分离菌对头孢噻肟、氟苯尼考、多西环素均敏感;OmpA基因进化分析显示,2株分离株的同源性100%,与鸡源、鸭源鸭疫里默氏菌参考株同源性分别为93.9%~94.0%,95.8%~99.9%;SPF鸡注射细菌后可导致鸡关节出现胶冻样物;病理学变化显示,肾小管上皮细胞水肿或变性,神经元水肿或皱缩.试验证明肉种鸡可以感染鸭疫里默氏菌,感染鸭疫里默氏菌后可能对鸡的生殖系统或腿部关节造成损伤.
To identify the two strains of similar pathogenic bacteria isolated from the caseous exudate in the oviduct of broiler breeders and the yolks of their hatching eggs,the two isolates were subjected to Gram staining,serotype identification,drug sensitivity test and OmpA gene analysis.Additionally,patho-genicity tests were conducted on SPF chickens using the isolated bacteria.The results showed that both strains isolated from the oviduct and egg yolk were identified as type 1 Riemerella anatipestifer.Both strains were found to be sensitive to cefotaxime,florfenicol and doxycycline.Phylogenetic analysis of the OmpA gene revealed 100%homology between the two strains,while the homology with reference strains of Riemerella anatipestifer from chickens and ducks ranged from 93.9%~94.0%and 95.8%~99.9%,re-spectively.Injecting bacteria into SPF chickens resulted in gelatinous samples in the joints of chickens.Pathological changes included tubular epithelial cell edema or degeneration and neuronal edema or shrink-age.The experiment showed that broiler chickens can be infected with Riemerella anatipestifer,which may lead to damage to the reproductive system or leg joints following infection.
