从内源性抗菌肽研究尿感方对膀胱上皮细胞感染早期防御响应的影响OA
Investigating the Effects of"Urinary-infection Prescription"on Early Defensive Response in Bladder Epithelial Cell Infection via Antimicrobial Peptides
目的:从内源性抗菌肽(antimicrobial Peptides,AMPs)的表达观察尿感方对膀胱上皮细胞(bladder epithelial cells,BECs)感染早期防御响应的影响,并从IL-6/Stat3信号通路探讨其作用机理.方法:采用RT-PCR法检测尿道致病性大肠杆菌感染早期BECs内源性抗菌肽CAMP、DEFB1、脂质运载蛋白2(Lipocalin 2,LCN2)、核糖核酸酶4(Ribonuclease 4,RNases4)和RNase7 mRNA的表达,并观察细菌定植率,同时采用ELISA法检测白细胞介素6(Interleukin-6,IL-6)水平,蛋白免疫印迹法检测p-Stat3蛋白表达.结果:与模型组比较,5%、10%大鼠空白尿液组AMPs(CAMP、DEFB1、LCN2、RNases4和Nases7)的mRNA表达、细菌定植率、IL-6水平和p-Stat3表达均无明显差异(P>0.05);与5%大鼠空白尿液组比较,5%大鼠含药尿液组Nases7的mRNA表达上调(P<0.05);与10%大鼠空白尿液组比较,10%大鼠含药尿液组AMPs(CAMP、DEFB1、LCN2、RNases4和Nases7)的mRNA表达、IL-6水平和p-Stat3表达均上调(P<0.05),细菌定植率下降(P<0.05).结论:尿感方能增加感染早期BECs的AMPs表达,强化BECs对病原菌的快速防御响应,减少细菌定植,其部分作用机制与IL-6/Stat3信号传导通路有关.
Objective:To observe the effects of urinary-infection prescription on early defensive response in bladder epithelial cells(BECs)infection via the expressions of AMPs,and to survey its mechanism from IL-6/Stat3 signaling pathway.Methods:RT-PCR was applied to detect the expressions of AMPs including CAMP,DEFB1,LCN2,RNases4 and RNase7 mRNA in BECs at early stage of uropathogenic escherichia coli(UPEC)infection,to observe bacterial colonization rate,meanwhile ELISA was used to measure the levels of IL-6,Western blotting to monitor the expressions of p-Stat3 protein.Results:No significant difference existed in the expressions of mRNA of AMPs including CAMP,DEFB1,LCN2,RNases4 and Nases7,bacterial colonization rate,the levels of IL-6 and p-Stat3 when 5%and 10%normal rat urine control groups were compared with the model group(P>0.05);the upregulation of Nases7 mRNA was observed when 5%drug-containing rat urine group was compared with 5%normal rat urine control group(P<0.05);10%drug-containing rat urine group demonstrated the increase in mRNA expressions of AMPs including CAMP,DEFB1,LCN2,RNases4 and Nases7,the levels of IL-6 and p-Stat3,while the decrease in bacterial colonization rate when compared with 10%normal rat urine control group(P<0.05).Conclusion:Urinary-infection prescription could enhance the expressions of AMPs in BECs during the early stage of infection,enhance rapid defense response of BECs against pathogenic bacteria,and reduce bacterial colonization rate,and part of the mechanism is related to IL-6/Stat3 signaling pathway.
