Functional Magnetic Resonance Imaging and Optical Imaging Integration in the Screening of High-risk Populations for Mental Disorders
目的:本研究旨在探讨功能性磁共振成像(functional magnetic resonance imaging,fMRI)与光学成像技术(functional near-infrared spectroscopy,fNIRS)集成在筛查精神疾病高风险人群中的有效性,为精神疾病的早期干预提供可靠的科学依据.方法:本研究招募了60名来自精神疾病家族史的高风险人群与60名健康对照人群,所有参与者均为18~35岁.通过fMRI评估脑区活动的空间分辨率,并结合fNIRS监测前额叶皮层氧合血红蛋白(oxygenated hemoglobin,HbO)浓度的变化,从而评估脑区活动的强度.使用功能连接强度分析(functional connectivity strength,FCS)、独立成分分析(independent component analysis,ICA)和小世界属性分析等方法,检测高风险个体在不同脑区间的连通性变化.结果:集成影像方法成功识别了高风险个体的特异性脑功能网络异常,尤其是默认模式网络(default mode network,DMN)和执行控制网络(executive control network,ECN)的连通性变化.与健康对照组相比,高风险组在认知任务中表现出前额叶皮层激活差异,以及双侧皮层HbO信号的同步性显著降低.结论:功能性磁共振成像与光学成像技术的集成在精神疾病高风险人群的早期筛查中展现了巨大的应用潜力,能够为精神疾病的早期干预提供可靠的依据.该方法的进一步发展可能为临床筛查和早期诊断提供有效工具.
Objective:This study aims to explore the effectiveness of integrating functional magnetic resonance imaging(fMRI)and functional near-infrared spectroscopy(fNIRS)in screening high-risk populations for mental disorders,providing a reliable scientific basis for the early intervention of these conditions.Methods:We recruited 60 participants from families with a history of mental disorders and 60 healthy controls,all aged 18~35.fMRI was used to assess the spatial resolution of brain activity,combined with fNIRS to monitor changes in the concentration of oxygenated hemoglobin(HbO)in the prefrontal cortex,thus evaluating the intensity of brain activity.Various methods,including functional connectivity strength(FCS)analysis,independent component analysis(ICA),and small-world property analysis,were used to detect changes in connectivity between different brain regions in high-risk individuals.Results:The integrated imaging approach successfully identified specific brain functional network abnormalities in high-risk individuals,particularly changes in connectivity within the default mode network(DMN)and executive control network(ECN).Compared to the healthy control group,the high-risk group displayed differences in prefrontal cortex activation during cognitive tasks,as well as a significant reduction in the synchronicity of bilateral cortical HbO signals.Conclusion:The integration of functional magnetic resonance imaging and optical imaging technology shows great potential in the early screening of high-risk populations for mental disorders,providing a reliable basis for early intervention.Further development of this method may offer effective tools for clinical screening and early diagnosis.
