云南部分地区羊芽囊原虫感染情况调查及亚型鉴定OA

为了解云南省绵羊和山羊芽囊原虫(Blastocystis)的感染情况和基因分型分布,于2023年7月至2024年1月从昆明市、红河州、楚雄州采集绵羊和山羊粪样,提取粪便基因组DNA,基于芽囊原虫小亚基核糖体RNA(SSU rRNA)基因进行PCR扩增,将阳性样本测序后,在NCBI上进行BLAST比对分析,鉴定虫种和基因亚型.使用MEGA 11.0软件以邻接法(NJ)构建系统进化树,使用SPSS 20.0软件利用 x2检验对羊芽囊原虫检出率影响因素进行统计学分析.共采集138份羊粪样,其中35份(25.36%)粪样扩增出芽囊原虫SSU rRNA基因片段.昆明市、楚雄州、红河州的检出率分别为30.00%、24.07%和16.67%;奶山羊、黑山羊、绵羊的检出率分别为30.00%、24.14%和15.00%.不同地区、品种、养殖模式间检出率差异不显著.序列比对分析显示,云南省三个地区检出的芽囊原虫基因亚型有ST1、ST10和ST14,3份阳性样品鉴定为ST1亚型,9份阳性样品鉴定为ST14亚型,23份阳性样品鉴定为ST10亚型,ST10亚型为优势亚型(23/35).进化分析显示,获得的ST1、ST10和ST14样品数据均与其代表的基因型参考序列聚在一支,拓扑结构与之前的研究结果一致.结果表明,云南省三个地区的绵羊和山羊芽囊原虫感染普遍,优势基因亚型为ST10,且存在人兽共患基因亚型ST1,提示有芽囊原虫人兽共患风险,为后续公共卫生监测及芽囊原虫病的进一步研究提供数据支持.

To investigate the infection and subtype distribution of Blastocystis sp.in sheep and goats in Yun-nan province,fecal samples were collected from sheep and goats in Kunming city,Honghe prefecture,and Chuxiong prefecture from July 2023 to January 2024.Genomic DNA was extracted from the fecal samples,and PCR amplification was performed based on the small subunit ribosomal RNA(SSU rRNA)gene of Blastocystis sp.After sequencing the positive samples,Blast alignment was performed on NCBI to identify the species and gene subtypes.Phylogenetic tree was constructed using neighbor-joining method(NJ)of MEGA 11.0 software.The factors affecting the detection rate of Blastocystis sp.from sheep and goats were analyzed by x2 test using SPSS 20.0 software.A total 138 of sheep and goats fecal samples were collected,and 35(25.36%)fecal samples were amplified with SSU rRNA gene fragments of Blastocystis sp.The de-tection rates in Kunming city,Chuxiong prefecture and Honghe prefecture were 30.00%,24.07%and 16.67%,respectively.The detection rates of dairy goats,black goats and sheep were 30.00%,24.14%and 15.00%,respectively.There was no significant difference in the detection rate among different regions,vari-eties and breeding patterns.Blastocystis sp.were determined as ST1,ST10 and ST14 subtypes based on se-quence comparison.Three positive samples were identified as ST1 subtype,nine positive samples were iden-tified as ST14 subtype,while 23 positive samples were identified as ST10 subtype.The subtype ST10 was considered as the predominant subtype(23/35).Phylogenetic tree analysis showed that samples of ST1,ST10 and ST14 subtypes were all gathered together with the branches of their reference sequences,respec-tively.The topological structure of phylogentic analysis is in accordance with the previous studies.Blasto-cystis infection in sheep and goats is prevalent in three regions of Yunnan province,the predominant gene subtype is ST10.The results of present study show potential possibility of zoonosis of Blastocystis origina-ted from the zoonotic gene subtype ST1,which provides data support for further public health detection and prevention and control of Blastocysticosis.
