中医药调控JAK2/STAT3信号通路治疗脊髓损伤的研究进展OA
Research progress in the regulation of JAK2/STAT3 signaling pathway by traditional Chinese medicine in the treatment of spinal cord injury
脊髓损伤(SCI)是医学一大难题,目前SCI的主要治疗方式是早期手术减压和西药糖皮质激素脉冲治疗,但其副作用较大,且这些治疗很难有效抑制神经炎症引起的继发性损伤,从而促进神经功能的恢复,中药因其副作用较小受到国内外学者的关注.现代研究表明,SCI的发生发展后酪氨酸蛋白激酶2(JAK2)/转录激活因子3(STAT3)信号通路的持续异常激活可导致SCI相关炎症因子表达上升.本文通过对国内外相关文献进行分析,对JAK2/STAT3信号通路在SCI发生发展中的作用进行总结,从中药单体化合物、中药复方、针灸等方面围绕中医药通过调控JAK2/STAT3信号通路干预SCI发生发展的作用机制进行综述,以期为SCI的临床研究与药物研制提供新思路.
Spinal cord injury(SCI)is a major challenge in medicine.The current primary treatments for SCI are early surgical decompression and glucocorticoid pulse therapy.However,these therapies have considerable side effects and are often ineffective in suppressing neuroinflammation-induced secondary damage,thereby limiting the recovery of neurological function.Traditional Chinese medicine(TCM)has attracted attention from researchers both domestically and abroad due to its relatively minor side effects.Modern studies have shown that sustained abnormal activation of the Janus kinase 2(JAK2)/signal transducer and activator of transcription 3(STAT3)signaling pathway after the occurrence and progression of SCI can lead to increased expression of inflammatory factors associated with the SCI.This article analyzes relevant literature from both domestic and international sources to summarize the role of the JAK2/STAT3 signaling pathway in the onset and progression of SCI.Furthermore,from the perspectives of single compounds in Chinese herbal,traditional herbal formulas,and acupuncture,this paper reviews the mechanisms by which TCM intervenes in the occurrence and development of SCI through the regulation of the JAK2/STAT3 signaling pathway,aiming to provide new insights for clinical research and drug development of SCI.
