CTRP12调控平滑肌细胞表型转化在腹主动脉瘤中的功能研究OA
Functional study of CTRP12 in regulating vascular smooth muscle cell pheno-typic switching during abdominal aortic aneurysm pathogenesis
目的 本研究旨在探索CTRP12在腹主动脉瘤(AAA)的作用及潜在分子机制,重点关注其对炎症因子、基质金属蛋白酶(MMPs)以及VSMC表型转化的影响.方法 采集AAA患者和健康志愿者的血清样本,采用ELISA检测血清中CTRP12的表达水平;构建ApoE-/-小鼠AAA模型,通过皮下微量缓释泵注射血管紧张素Ⅱ(Ang Ⅱ)诱导AAA形成,采用尾静脉注射腺相关病毒干预CTRP12表达;通过组织染色观察腹主动脉组织病理变化;使用qPCR和Western blot分析AAA小鼠腹主动脉中CTRP12、炎症因子(IL-6、IL-1β、TNF-α)、基质金属蛋白酶(MMP2、MMP9)和VSMC表型标志基因的表达变化.结果 AAA患者的血清中CTRP12的表达显著升高;ApoE-/-模型小鼠的血清及腹主动脉组织中CTRP12的表达显著升高,IL-6、IL-1β和TNF-α浓度增加,MMP2和MMP9表达上调,通过干扰CTRP12的表达后,IL-6、IL-1β、和TNF-α浓度降低,MMP2和MMP9表达下调,抑制VSMC的表型转化.结论 CTRP12可能通过调控炎症反应、MMPs表达及VSMC表型转化参与AAA的病理进程.
Objective This study aimed to investigate the role of C1q/TNF-related protein-12(CTRP12)in Ab-dominal Aortic Aneurysm(AAA)and its underlying molecular mechanisms,with a focus on its effects on inflam-matory factors,matrix metalloproteinases(MMPs),and VSMC phenotypic switching.Methods Serum samples were collected from AAA patients and healthy volunteers,and CTRP12 levels were measured by ELISA.An AAA model was established in ApoE-/-mice via Angiotensin Ⅱ(Ang Ⅱ)infusion using osmotic minipumps.Adeno-associated virus was administered via tail vein injection to modulate CTRP12 expression.Pathological changes in the abdominal aorta were assessed by hematoxylin-eosin(HE)and Masson staining.The expression levels of CTRP12,inflammatory factors(IL-6,IL-1β,TNF-α),MMPs(MMP2,MMP9),and VSMC pheno-typic markers in the abdominal aorta of AAA mice were analyzed using qPCR and Western blot.Results CTRP12 expression was significantly elevated in the serum of AAA patients.In ApoE-/-mice,CTRP12 expression was markedly increased in both serum and abdominal aortic tissues,accompanied by elevated concentrations of IL-6,IL-1β,and TNF-α,and upregulation of MMP2 and MMP9.Knockdown of CTRP12 expression led to reduced levels of these inflammatory factors,downregulated MMP2 and MMP9 expression,and inhibited VSMC phenotyp-ic switching.Conclusion CTRP12 may accelerate AAA progression by regulating inflammatory responses,MMPs expression,and VSMC phenotypic switching.
