目的 探究中央杏仁核(CeA)中异氟烷激活的细胞及其与促肾上腺皮质激素释放激素(CRH)神经元的关系.方法 选用8只健康雄性CRH-tdTomato-Cre小鼠,随机分为异氟烷组(1.4%异氟烷麻醉2 h,n=4)和空气对照组(空气2 h,n=4).实验结束后,进行心脏灌注取脑,制备CeA脑区冰冻切片,随后行免疫荧光染色,通过荧光显微镜观察和图像采集.统计分析CeA各层面中被异氟烷激活的细胞数量及CRH神经元的分布和共定位情况.结果 与空气组相比,异氟烷组小鼠CeA脑区神经元活性显著增强,c-Fos阳性细胞数显著增加(P<0.05);被异氟烷激活的细胞中,部分为CRH神经元,但共定位的比例较低;被激活的细胞主要分布在CeA的靠后层面,而CRH神经元主要分布于CeA的靠前层面.结论 异氟烷可显著激活CeA脑区神经元,但其中仅有少部分为CRH神经元,大部分被激活细胞并非CRH神经元.

Objective The aim of this study was to investigate the isoflurane-activated cells in the central amyg-dala(CeA)and their relationship with corticotropin-releasing hormone(CRH)neurons.Methods Eight healthy male CRH-tdTomato-Cre mice were randomly assigned to two groups:the isoflurane group(1.4%isoflurane an-esthesia for 2 hours,n=4)and the air control group(n=4).After the experiment,cardiac perfusion was per-formed to collect the brains,and frozen sections of the CeA region were prepared.Immunofluorescence staining was then carried out,and fluorescence microscopy was used for observation and image acquisition.The number of isoflurane-activated cells and the distribution and co-localization of CRH neurons in different layers of the CeA were statistically analyzed.Results Compared to the air control group,the isoflurane group showed a significant increase in neuronal activity in the CeA,with a notable increase in c-Fos-positive cells(P<0.05).Some of the isoflurane-activated cells were CRH neurons,but the co-localization rate was low.The activated cells were pri-marily located in the posterior regions of the CeA,while CRH neurons were predominantly located in the anterior regions of the CeA.Conclusion Isoflurane significantly activates neurons in the CeA,but only a small proportion of these activated cells are CRH neurons,with most of the activated cells not being CRH neurons.
