目的:探究三七全株不同部位蛋白质在其对应药材鉴别时的应用价值.方法:选取三七地下部分(主根、筋条、剪口)和地上部分(茎叶、花)的鲜品和干品作为研究对象,提取总蛋白质,采用十二烷基磺酸钠-聚丙烯酰胺凝胶电泳进行蛋白质分离,结合液相色谱-质谱法筛选出具有多维度识别潜力的候选蛋白质.结果:三七全株不同药用部位鲜品的总粗蛋白质提取率均高于对应部位的市场药材干品,部分差异无统计学意义;仅文山与曲靖的三七(主根)鲜品、三七茎叶鲜品差异有统计学意义(P<0.05),其他药用部位的产地间差异无统计学意义;三七地下部分与地上部分鲜品的蛋白质电泳图谱存在明显差异.三七地下部分的鲜品和干品均以 60、25~35 kDa为主要特征条带,而三七地上部分的鲜品和干品均无此特征条带.三七地下部分干品药材的特征条带与同属人参药材的带型相似.液相色谱-质谱法分析结果显示,三七地下部分与人参药材在 60、25~35 kDa处共有的 2 个特征条带区域均以核糖核酸酶样储存蛋白为主;进一步的氨基酸序列比对结果表明,与人参比较,三七地下部分的核糖核酸酶样储存蛋白氨基酸序列有 6 个差异位点.结论:可通过总蛋白质电泳图谱鉴别三七地下部分与地上部分,特征条带主要成分为核糖核酸酶样蛋白.

Objective:To explore the application value of proteins from different medicinal parts of Panax notoginseng for the identification of corresponding medicinal materials.Methods:The total crude protein was extracted from the fresh and dried samples of underground parts(main root,rootlets,and rhizomes)and aboveground parts(stems and leaves,flowers)and separated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis(SDS-PAGE).The protein candidates for multidimensional identification were selected by mass spectrometry combined with liquid chromatography.Results:The extraction rate of total crude protein from fresh medicinal parts of P.notoginseng was higher than that from corresponding dried medicinal parts,with some non-significant differences.The fresh samples showed no significant differences among different producing areas,except the main root and the stems and leaves of P.notoginseng from Wenshan and Qujing(P<0.05).There were significant differences in the protein electrophoresis profiles of fresh samples between the underground and aboveground parts.The bands at 60 kDa and 25~35 kDa were specific to fresh and dried underground parts,while they were absent for fresh or dried aboveground parts.The specific protein bands of underground parts from P.notoginseng were similar to those of P.ginseng.The results of mass spectrometry combined with liquid chromatography showed that the two specific protein bands from P.notoginseng and P.ginseng were RNase-like storage proteins.Moreover,the amino acid sequence comparison of RNase-like storage proteins from the underground parts of P.notoginseng and P.ginseng showed 6 varied amino acid sites.Conclusion:The underground and aboveground parts of P.notoginseng can be identified by the protein profile,and the significant characteristic bands are mainly RNase-like proteins.
